Soluble thrombomodulin attenuates sinusoidal obstruction syndrome in rat through suppression of high mobility group box 1

Abstract: rTM preserved SECs and attenuated MCT-induced SOS in rats through suppression of circulatory HMGB1 and neutrophil accumulation, resulting in improved survival after hepatectomy.

“…• HMGB1 acts as a mediator playing a key role both in the early and late stages of systemic inflammation and its acetylated form may be a better biomarker for prognosis prediction in DILI. “…The treatment protocol followed the rat SOS model with some modifications. 20 Briefly, MCT (200 mg per kg body weight) was administered to mice via IP injection on day 1 to induce SOS. Concurrently, either TME5 (1 mg per kg body weight, n = 6) or control diluent (double distilled water, n = 6) was administered to these mice via IP injection on days 1 and 2.…”

“…20 They demonstrated that administration of MCT to rats increases serum levels of HMGB1 and non-histone nuclear proteins, and that rTM effectively counteracts the MCT-mediated increase in levels of HMGB1. HMGB1 is released from necrotic cells as well as activated inflammatory cells and stimulates further production of inflammatory cytokines in association with activation of NF-κB.…”

“…Over the last years, evidence from animal experiments is emerging that ART‐123 has important organ protective effects and that it has cytoprotective effects on the endothelium . In a rodent model of hepatic warm ischemia, livers that were ex vivo perfused with ART‐123, after 6 hours of static cold preservation, showed significantly improved bile production and decreased sinusoidal narrowing compared with controls .…”
