Soluble B7-H1: Differences in production between dendritic cells and T cells

Abstract: Tumor cells aberrantly express several T cell inhibitory molecules including members of the B7-H co-regulatory family. Presumably tumor-expressed B7-H1 and B7-H3 confer resistance to elimination by the immune system. In addition, elevated levels of soluble B7-H1 (sB7-H1) has been identified in the sera of cancer patients, including renal carcinoma patients and is associated with increased cancer related death. Here we report that sB7-H1 is produced and released by activated mature dendritic cells (mDC). Immatu… Show more

“…However, the underlying pathogenesis remains unclear. Amarnath et al ( Although both myeloid and T lymphocytes exhibit mPD-L1 expression, the release of sPD-L1 is a feature of the myeloid population (32,33). The results of the present study demonstrated that the expression levels of sPD-L1 in PF were increased, as compared with PB; this may be due to the high concentrations of cytokines in the local pleural cavity.…”

“…The results of Fig. 4C shows that DNA-PKcs and B7-H1 in Karpas 299 were mutually pulled down by their respective antibodies20. Significantly, an association of DNA-PKcs and B7-H1 was detected in activated (known to up-regulate B7-H1 expression) but not resting human T cells (Fig.…”

“…24,25 It has also been shown that sPD-L1 is produced and released by activated mature dendritic cells and suggested that immune and tumour cells could be the source of soluble PD-L1. 26 Measuring sPD-L1 may be a surrogate marker for surface PD-L1, which could be readily tested in the clinic with a blood test. Further work is required to explore whether these immunological markers in blood are raised in other conditions of inflammation, chronic infection and autoimmunity to establish their efficacy as diagnostic biomarkers and also to monitor response to therapy or recurrence of disease.…”
