Solubilization of Poorly Water-Soluble Drugs Using Solid Dispersions

Abstract: Many new drugs have been discovered in pharmaceutical industry and exposed their surprised potential therapeutic effects. Unfortunately, these drugs possess low absorption and bioavailability since their solubility limitation in water. Solid dispersion (SD) is the current technique gaining so many attractions from scientists due to its effect on improving solubility and dissolution rate of poorly water-soluble drugs. A number of patents including the most recent inventions have been undertaken in this review t… Show more

“…Other common instrumental analyses are recommended for various purposes of a study. For instance, to detect drug crystallinity in a dried FFS, powder X-ray diffraction and differential scanning calorimetry are critical analyses; or to identify the molecular interactions between the drug and other excipients (or between excipients), Fourier-transformed infrared spectroscopy and nuclear magnetic resonance should be included [27,76,140,141,142].…”

“…NMR is another powerful technique that can be used to perform high sensitivity detection and obtain quantitative information at the molecular level [ 87 ]. In general, NMR monitors possible changes of electron density around specific interacting atoms to detect molecular interactions at the atomic level [ 88 , 89 ].…”

“…While an alternative structure of drug from crystalline to amorphous state may occur to improve the dissolution, an interaction among agents is another factor to contribute to the enhanced drug dissolution. Although molecular interaction between drug and polymer has been known as an important factor for improving dissolution rate [3,7,[16][17][18], there have been no studies through sonoprecipitation method indicating modulation of molecular interactions and its interesting effects on particle size for the control of drug dissolution rate in details.…”
