Solid Dispersions: Revival with Greater Possibilities and Applications in Oral Drug Delivery

Abstract: Improvement of oral bioavailability of poorly water-soluble drugs remains one of the most challenging aspects of drug development. Solid dispersions seem to be a viable technique for overcoming this problem. However, the practical applicability of these systems has remained limited because of difficulties in conventional methods of preparation, poor reproducibility of physicochemical properties, difficulties in dosage form development, and lack of feasibility for scale-up of manufacturing processes. This revie… Show more

“…[27] However, the practical applicability of solid dispersion systems has remained limited due to difficulties in conventional methods of preparation, poor reproducibility of physiochemical properties, dosage formulation and lack of feasibility for scaling-up manufacturing processes. [28] Electrospun nanofibers may provide novel approaches as to how the dissolution rate of even very poorly soluble compounds might be improved to minimize the limitations of oral availability.…”

“…The SD technique produces a significant increase in surface area and surface wettability as well as solid state modification from crystalline to amorphous form. However, the SD characteristics can be influenced by several factors such as preparation method, carrier type and drug:carrier ratio and pH modifiers [7,8].…”
