Microbial functional diversity is significantly associated with both nutrient cycling and organic matter decomposition. However, how different forests as well as the soil parent materials influence the soil microbial carbon metabolism remains poorly understood. In this study, a natural secondary forest and a Pinus yunnanensis plantation, with similar climatic conditions under contrasting parent materials (clasolite in the non-karst areas and limestone in the karst areas) in Yunnan Province, China, were investigated. The soil microbial carbon metabolism diversity was assessed by the Biolog® ECO-plates. During the dry season, the soil microbial communities used carbon substrate in secondary forest and Pinus yunnanensis plantation, showing no significant difference, both in non-karst and karst areas. The microbial communities in the non-karst area were more efficient in utilizing carbon substrates than those in the karst area with the same vegetation types, resulting in the higher accumulation of organic carbon in the karst area. The six categories of most frequently utilized carbon substrates were carbohydrates, carboxylic acids, and amino acids in both the non-karst and the karst areas. The soil basal respiration of the secondary forest was higher than that of the Pinus yunnanensis plantation, both in the non-karst and the karst areas. In addition, the driving factors of the soil microbial community functional diversity in the non-karst and karst areas are different. Our findings suggest that soil microbial functional diversity is governed by vegetation types as well as by soil properties in subtropical forests. Moreover, calcareous soil holds a higher proportion of recalcitrant organic carbon, which is difficult to utilize by microorganisms.

The effects of interactions of soil type and soil depth on soil C:N:P stoichiometry and microorganisms are poorly understood. In this study, soil samples (0–10, 10–20, 20–30, 30–50, 50–100 cm) were collected from two soil types (Haplic luvisols and Eutric cambisols) in Sabina przewalskii Kom. forest of Qinghai-Tibet Plateau. The soil C:N ratio and soil microbial biomass (SMB) measured using phospholipid fatty acid in Eutric cambisols were significantly higher than in Haplic luvisols, while soil C:P and N:P ratios were the opposite. In the two soil types, the soil C:N ratio significantly increased with soil depth, and the soil C:P and N:P ratios declined. Structural equation modelling (SEM) indicated that soil depth directly affected soil C, N and P contents. Soil type and soil depth could directly affect soil fungal and bacterial biomass, and indirectly affect both of them through soil bulk density. Meanwhile soil fungal biomass was influenced by soil depth through Total C. These results highlighted that the vertical distribution of soil bacteria could largely be attributed to changes of soil fungi depending on soil carbon resources.

Ants are important soil engineers, affecting the structure and function of ecosystems. To address the impacts of ants (Camponotus herculeanus) on the properties of an alpine meadow ecosystem of Qinghai–Tibet Plateau, we investigated the effects of ant mounds on plant biomass, soil physicochemical properties, microbial diversity, and functions. We found that the total biomass of plant community was significantly greater in ant mound periphery. Plant species richness in ant mounds was reduced compared with that of control plots without ant mounds. Significant changes in physicochemical properties of soil were also observed. Soil organic matter, total nitrogen, available phosphorous, total potassium, and available potassium increased in ant mound soil due to the excavation activities by ants as well as the accumulation of organic matter and other nutrients during mound construction. For example, roots/soil contents (g/g) and soil moisture in ant mound soils were lower than those in controls. Microbial community composition and microbial biomass were clearly changed in ant mound soils. BIOLOG analysis further indicated that the functional diversity of the microbial community of ant mound soil increased and differed from that of controls. This study indicates that ant‐induced modification of soil properties indirectly influences plant biomass and species composition, and ant mounds have different microbial communities from those of control soil.
