Although irrigation water is frequently assessed for the presence of plant pathogens, large spatial and temporal surveys that provide clues on the diversity and circulation of pathogens is missing. We evaluate the diversity of soft rot Pectobacteriaceae (SRP) of the genera Dickeya and Pectobacterium over two years in a temperate, mixed use watershed. The abundance of isolated strains correlates with the agricultural gradient along the watershed with a positive correlation found with temperature, nitrate and dissolved organic carbon water concentration. We characterized 582 strains by amplification and sequencing of the gapA gene. MLSA analysis performed with 3 housekeeping genes for 99 strains and core genome analysis of 38 sequenced strains confirmed for all the strains but one the taxonomic assignation obtained with the sole gapA sequence. Pectobacterium spp. (549 isolates) were far more abundant than Dickeya spp. (33 isolates). Dickeya spp. were only observed in the lower part of the river when water temperature was above 19°C and we experimentally confirmed a decreased fitness of several Dickeya spp. at 8°C in river water. D. oryzae dominates the Dickeya spp. P. versatile and P. aquaticum dominate the Pectobacterium spp. but their repartition along the watershed was different, P. versatile being the only species regularly recovered all along the watershed. Excepting P. versatile, Dickeya and Pectobacterium spp. responsible for disease outbreak on crops were less abundant or rarely detected. This work sheds light on the various ecological behaviours of different SRP in stream water and indicates that SRP occupation is geographically structured.

To the present day, no efficient plant protection method against economically important bacterial phytopathogens from the Pectobacteriaceae family has been implemented into agricultural practice. In this view, we have performed a multivariate optimization of the operating parameters of the reaction-discharge system, employing direct current atmospheric pressure glow discharge, generated in contact with a flowing liquid cathode (FLC-dc-APGD), for the production of a plasma-activated liquid (PAL) of defined physicochemical and anti-phytopathogenic properties. As a result, the effect of the operating parameters on the conductivity of PAL acquired under these conditions was assessed. The revealed optimal operating conditions, under which the PAL of the highest conductivity was obtained, were as follows: flow rate of the solution equaled 2.0 mL min−1, the discharge current was 30 mA, and the inorganic salt concentration (ammonium nitrate, NH4NO3) in the solution turned out to be 0.50% (m/w). The developed PAL exhibited bacteriostatic and bactericidal properties toward Dickeya solani IFB0099 and Pectobacterium atrosepticum IFB5103 strains, with minimal inhibitory and minimal bactericidal concentrations equaling 25%. After 24 h exposure to 25% PAL, 100% (1−2 × 106) of D. solani and P. atrosepticum cells lost viability. We attributed the antibacterial properties of PAL to the presence of deeply penetrating, reactive oxygen and nitrogen species (RONS), which were, in this case, OH, O, O3, H2O2, HO2, NH, N2, N2+, NO2−, NO3−, and NH4+. Putatively, the generated low-cost, eco-friendly, easy-to-store, and transport PAL, exhibiting the required antibacterial and physicochemical properties, may find numerous applications in the plant protection sector.

The article discusses the morphological changes of birch trees due to bacterial dropsy growing in the "Green Belt" artificial forest in the surrounding of the Nur-Sultan city. Samples of kernel were taken from diseased tree trunks, and, afterward, isolated bacterial cultures were grown in a nutrient medium. For molecular genetic studies, one strain (designated as DDdd-1), the most typical of morphological characteristics, was selected, the colonies of which showed the greatest growth on nutrient media. The nucleotide sequences 8F and 806R of 16SrRNA were determined on the basis of sequencing. The resulting sequences were compared in GenBank from the NCBI database showing related species and their similarities. Extraction of cores from infected birch trunks, isolation of pure strains of the pathogen, molecular identification of ribosomal RNA 16S nucleotide chains were performed using modern methods in the field of bacteriology. Then, the results obtained were compared with relevant data from the International Genbank database. Molecular characteristics of isolated bacterial DDdd-1 strain have been found to correspond to typical Dickeya dadantii by 96.72%. According to the International Genbank, the DDdd-1 strain is defined as pathovar D. dadantii pv. betulae. The data obtained demonstrate close relation of DDdd-1 strain Dickeya dadantii pv. betulae to some strains of Dickeya chrysanthemi. The species identified in International Genbank as Erwinia chrysanthemi with index numbers KJ541470.1 and AF373175.1 are assumed to be identical to D. dadantii pv. betulae. Dickeya is a complex taxon that still requires a lot of research efforts on species composition, close family bonds, the size and structure of host plants.
