Sodium New Houttuyfonate Affects Transcriptome and Virulence Factors of Pseudomonas aeruginosa Controlled by Quorum Sensing

Abstract: As a major opportunistic pathogen, Pseudomonas aeruginosa can produce various virulence factors and form biofilms. These processes are controlled by the quorum sensing (QS) system. Sodium new houttuyfonate (SNH) is an adduct of houttuyfonate, the main component of the common Chinese medicine plant Houttuynia cordata, which has antibacterial and anti-inflammatory effects. We evaluated the effect of SNH on P. aeruginosa biofilms, virulence factors, and transcription.… Show more

“…Hence, QS is a critical target for the design of novel antimicrobial therapeutics. Some TCMMs have been shown to inhibit QS in addition to their antimicrobial activity Frontiers in Pharmacology frontiersin.org (Lee et al, 2014;Chen et al, 2016;Zhao et al, 2020). In this study, the QS inhibitory activity of thirty-four TCMMs was investigated by biosensor CV026.…”

“…Structurally, SNH is more stable than houttuynin [ 42 ]. Currently, SNH is an antibacterial agent mainly for inflammatory diseases, such as chronic bronchitis, pneumonia, and other respiratory diseases, which is effective for infections caused by Staphylococcus aureus , Streptococcus pneumoniae, Streptococcus mutans, Pseudomonas aeruginosa and Candida albicans [ 34 , 43 , 44 , 45 , 46 ]. MET, as a common clinical antibacterial agent against anaerobic bacteria, is widely used in the treatment of infectious diseases.…”

“…In a previous study, sodium houttuyfonate effectively inhibited the synthetic lasl gene of AHL and reduced the level of expression, and the AHL receptor and the transcriptional regulator LasR were also inhibited, thereby downregulating the expression of the virulence factors pyocyanin and LasA; therefore, the PA population size can be effectively controlled through AHL mediation (Wu et al, 2014). Moreover, in a treatment evaluation of sodium houttuyfonate, the expression of the rhll and pqsA genes, which play key roles in the QS system, was significantly reduced and interfered with the production of pyocyanin; additionally, biofilm formation was monitored, and with the exception of lasA, the expression of the LasB, LecA, phzM, pqsA, and pilG genes was affected, which further inhibited the activation of PA virulence factors and biofilm formation (Zhao et al, 2020). These research results provide new insights for anti-PA activity.…”
