Sodium Butyrate Inhibits the Inflammation of Lipopolysaccharide-Induced Acute Lung Injury in Mice by Regulating the Toll-Like Receptor 4/Nuclear Factor κB Signaling Pathway

Abstract: Bacterial pneumonia is a common disease in dairy herds worldwide, which brings great economic losses to farmers. Sodium butyrate (SB), an inhibitor of histone deacetylase, plays an important role in limiting inflammation. The purpose of this study was to investigate the protective effects of SB on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice and explore the potential mechanism of SB protection. A total of 30 ICR mice were randomly divided into three groups (n = 10): a phosphate-buffered sal… Show more

“…TLR4 presents in the surface of monocytes/macrophages and it induces an innate immune response in mammals by recognizing LPS. Once LPS is recognized by TLR4, it leads to activate signal transduction and then induces NF-kB activation (Liu et al, 2019a). NF-kB is a ubiquitous transcription factor and closely related to the transcription of the genes responsible for the expression of pro-inflammatory cytokines (Hoffmann et al, 2002).…”

“…118 Sodium butyrate significantly decreased the TLR4 and NFKB signaling pathways in lipopolysaccharide-induced acute lung injury in mice. 119 In addition, butyrate inhibits the NF-kB pathway in the HT-29 colon cancer cell line. 120 However, this effect might be tissue-or cell type-specific.…”

“…LPS can cause severe pneumonia and sepsis and activate the downstream nuclear factor-kappa B (NF-κB) signaling pathway producing inflammatory mediators [7,8]. LPS was widely used in preparation of ALI animal models [9][10][11][12]. Among pulmonary cells, vascular endothelial cells (ECs) perform the important barrier between intravascular (IV) and extravascular (EV), which are always the primary target of inflammatory agents in local or systemic inflammation.…”
