SNP in Potentially Defunct Tetrahydrocannabinolic Acid Synthase Is a Marker for Cannabigerolic Acid Dominance in Cannabis sativa L.

Abstract: The regulation of cannabinoid synthesis in Cannabis sativa is of increasing research interest as restrictions around the globe loosen to allow the plant’s legal cultivation. Of the major cannabinoids, the regulation of cannabigerolic acid (CBGA) production is the least understood. The purpose of this study was to elucidate the inheritance of CBGA dominance in C. sativa and describe a marker related to this chemotype. We produced two crossing populations, one between a CBGA dominant cultivar and a tetrahydrocan… Show more

“…C. sativa inbred lines ‘TS1-3’ and ‘P163’ were used as parents in all experiments; both are female inbred lines developed by Oregon CBD (Independence, Oregon). Additional information on ‘TS1-3’ is available in Garfinkel et al [ 25 ]. Diploid F 1 hybrid seed of these parents has been released as ‘Stem Cell CBG’ and triploid F 1 hybrid seed has been released as ‘Stem Cell CBG Seedless’.…”

“…Analyses of cannabinoid content of the inflorescences were performed according to a modified protocol described by Vaclavik et al [ 27 ]; details regarding the modifications can be found in Garfinkel et al [ 25 ]. Five to 10 randomly chosen dried inflorescences from each plant were collected and used in the analysis.…”

“…Thus, harvesting hemp very early yields higher levels of CBGA compared to later in the plant’s life. In addition, some cultivars have increased yields of CBGA through selective breeding to inhibit its transformation into other cannabinoids during the plant’s maturation ( Garfinkel, et al, 2021 ). Similar to CBDA, CBGA displays low affinity for both the CB1 and CB2 receptors ( Navarro, et al, 2020 ).…”

“…This is probably due to a single amino acid mutation leading to a defective (B T0 ) variant ( Onofri et al 2015 ). Recently, a different CBGA-dominant cultivar was found to have another single amino acid mutation in THCAS ( Garfinkel et al 2021 ). Gene copy number variation has been suggested based on amplicon sequencing of the THCAS locus ( McKernan et al 2015 ; Weiblen et al 2015 ; Vergara et al 2019 ).…”
