Slug-Dependent Upregulation of L1CAM Is Responsible for the Increased Invasion Potential of Pancreatic Cancer Cells following Long-Term 5-FU Treatment

Abstract: BackgroundPancreatic adenocarcinoma is a lethal disease with 5-year survival of less than 5%. 5-fluorouracil (5-FU) is a principal first-line therapy, but treatment only extends survival modestly and is seldom curative. Drug resistance and disease recurrence is typical and there is a pressing need to overcome this. To investigate acquired 5-FU resistance in pancreatic adenocarcinoma, we established chemoresistant monoclonal cell lines from the Panc 03.27 cell line by long-term exposure to increasing doses of 5… Show more

“…The appearance of EMT features in 5-FU-resistant cells in vitro has been reported in various solid tumours, including PDAC,49 50 51 but a cause–effect relationship is lacking. Modulation of GATA6 levels in TKCC cells did not change their sensitivity to chemotherapeutic drugs, suggesting that GATA6 is part of a molecular phenotype involved in drug response, but it is not its major driver.…”

“…The TWIST1 target gene Slug has been shown to regulate L1CAM expression in cancer3839. Moreover, L1CAM has been linked to resistance to apoptosis in both intestinal tissue and an EOC cell line, and to drug resistance in pancreatic cancer404142.…”

“…A disadvantage of this strategy is that such clones in general are not representative for the entire resistant population. Therefore, three individual clones were selected through limited dilution after creation of stable, proliferating 5-FU resistant cells (Lund et al, 2015).…”
