Various methods have been used to isolate tissues or cell types from animals in order to determine the sites of plasma protein formation. Studies on the effect of extirpation of (1, 2) and of damage to (3-6) liver and spleen have indicated the role of these organs, but more specific evidence has been obtained with the use of perfused isolated organs (7-9) and of tissue culture (10-19).The results of such studies have shown that the liver is the main, if not the only site of albumin formation; and moreover, that most of the o~-and some of the fl-globulins are formed in the liver. The formation of clotting factors (16) and fibrinogen (7,13) by the liver was also demonstrated; and the studies of Rice el al. (20,21) indicate

The site of formation of C-reactive protein (CxRP, CRP) has been studied with tissues from rabbits, monkeys, and human beings. Rabbits and monkeys were stimulated to produce the acute phase protein by injection of turpentine, croton oil, endotoxin, paratyphoid-typhoid vaccine, or pneumococci. C14-amino acid incorporation in vitro was demonstrated by means of autoradiography of immunoelectrophoretic patterns made with culture fluids. It was found that among many different tissues tested liver was the only tissue which incorporated C14-lysine and isoleucine into CxRP or CRP. Only livers taken 16 to 24 hr after various stimuli were active; livers from normal animals or from animals killed 3 to 9 hr after stimulation did not produce detectable amounts of CxRP. Inflamed muscle from the injection site did not show C14-amino acid incorporation into CxRP. Several human tissues were also cultured, and a few liver cultures found to contain labeled CRP. The formation of CxRP or CRP by the liver was always accompanied by enhanced C14-amino acid incorporation into other serum proteins, but the reverse was not always found.

\s=b\This report describes the clinical, biochemical, and hepatic morphologic findings in ten children with severe serum \g=a\1-antitrypsin deficiency. Genetic protease inhibitor (Pi) phenotyping, using acid-starch gel and crossed antigen-antibody electrophoresis, demonstrated Pi phenotype ZZ in all our cases. In eight patients, manifestations of liver disease appeared during the first year of life. The case reports show that \g=a\1-antitrypsin deficiency should be suspected in any child with neonatal hepatitis, unexplained hepatomegaly or splenomegaly, or cirrhosis.In our report, one infant is normal at age 6 months, and one infant had progressive hepatic damage that culminated in liver failure and death at age 6 months. The variable clinical course and prognosis for infants with severe \g=a\1-antitrypsin deficiency is well illustrated by these two infants.
