Site-Specific Carboxypeptidase B1 Tyrosine Nitration and Pathophysiological Implications following Its Physical Association with Nitric Oxide Synthase-3 in Experimental Sepsis

Abstract: LPS-induced sepsis results in oxidative modification and inactivation of carboxypeptidase B1 (CPB1). In this study, immunoprecipitated CPB1 was probed for tyrosine nitration using monoclonal nitrotyrosine-specific Abs in a murine model of LPS-induced sepsis. Tyrosine nitration of CPB1 was significantly reduced in the presence of NO synthase (NOS) inhibitors and the xanthine oxidase (XO) inhibitor allopurinol and in NOS-3 knockout (KO) mice. CPB1 tyrosine nitration and loss of activity by the concerted action o… Show more

“…Tyrosine nitration is a marker for tyrosine radical formation (3234) The immunoreactivity of 3-nitrotyrosine could be localized in both the sinusoidal spaces and hepatocytes of the liver of CCl 4 -treated DIO, high fat-fed p47 phox and P2X7 receptor knockout mice (Fig. 4B i), and mean fluorescence intensities from nitrotyrosine immunoreactivity were significantly higher in CCl 4 -treated DIO mice than in untreated DIO mice (P<0.05; Fig.…”

“…The formation of nitrotyrosine in cellular proteins has been associated with modifications of protein activities (Curtis et al, 1996;Chatterjee et al, 2009). Furthermore, × NO has been reported to react with the tyrosyl radical of ribonucleotide reductase, resulting in the inhibition of DNA synthesis and ultimately cell death (Lepoivre et al, 1994).…”

“…This suggests that endothelial cells latently infected with KSHV might be inhibited in their ability to induce neutrophil chemotaxis. Therefore, we tested the effects of KSHV infection or LANA-1 expression on neutrophil chemotaxis using a modified Boyden chamber assay (17). Neutrophils were placed in the top wells of the chamber to migrate into bottom wells.…”
