SIRT4 and SIRT6 Serve as Novel Prognostic Biomarkers With Competitive Functions in Serous Ovarian Cancer

Abstract: Sirtuins (SIRTs) are class III histone deacetylases (HDACs) that include seven members and are widely expressed in mammals. Accumulating evidence shows that sirtuins may have contradictory roles in various malignancies. They mainly participate in metabolic homeostasis, DNA damage repair, cell survival, and differentiation, as well as other cancer-related biological processes. To better understand their prognostic role and biological functions, we used comprehensive bioinformatic analyses to demonstrate the exp… Show more

“…Abnormal SIRT6 expression is associated with a variety of malignant tumors, such as gastric cancer (48), ovarian cancer (49), medulloblastoma (50), etc. There is also evidence that SIRT6 can be regarded as an effective tumor suppressor owing to its effective regulation of aerobic glycolysis of cancer cells, which suppresses tumor metabolism (17).…”

“…Sirtuins have garnered significant attention for their involvement in cancer development and their potential as targets for new therapeutic interventions. This is because sirtuins play a crucial role in regulating metabolic reprogramming, modifying the cancer microenvironment, and maintaining genome stability [76][77][78][79]. Sirtuins are a family of evolutionarily conservative NAD + -dependent proteins, the third class of histone deacetylases with deacetylase or ADP-ribosyltransferase activity.…”

“…SIRT6 was found to be overexpressed in skin cancer and non-small cell lung cancer (NSCLC) [ 168 , 169 ], osteosarcoma, colon carcinoma, serous ovarian cancer, and clear cell renal cell carcinomas (ccRCC) [ 169 , 170 , 171 , 172 , 173 ], showing a poor prognostic value. However, in certain types of cancer, SIRT6 is classified as a tumor suppressor.…”
