SIRT3 deficiency delays diabetic skin wound healing via oxidative stress and necroptosis enhancement

Abstract: Sirtuin 3 (SIRT3) plays a vital role in several dermatological diseases. However, the role and detailed mechanism of SIRT3 in diabetic wound healing are unknown well yet. To explore possible involvement of SIRT3 and necroptosis in diabetic skin wound healing, SIRT3 knockout (KO) mice and 129S1/SvImJ wild‐type (WT) mice were injected with streptozotocin (STZ), and mice skin fibroblasts were exposed to high glucose (HG). It was found that SIRT3 expression decreased in the skin of diabetic patients. SIRT3 deficie… Show more

“…H 2 S also attenuated hydrogen peroxide-induced NLR family pyrin domain-containing protein 3 (NLRP3) inflammasome activation via SIRT3 in macrophages [45]. Moreover, we verified that SIRT3 deficiency exacerbated diabetic cardiomyopathy and delayed diabetic skin wound healing [46,47]. In our present experiment, mRNA expression of SIRT1 and SIRT3 was decreased, while other subtypes of the SIR2 family did not change significantly in cardiac fibroblasts with Ang II stimulation.…”

“…It serves to regulate the mitochondrial function, which is involved in regulating cellular redox status, mitochondrial energetics, biogenesis, dynamics, and apoptosis. Recent work has demonstrated that the lack of SIRT3 reduced blood supply, inhibited vascular endothelial growth factor (VEGF) expression, promoted superoxide production, impaired total antioxidant capacity, and consequently resulted in delayed skin wound healing in diabetic mice [ 22 ]. Therefore, sirtuins may be a new therapeutic target to improve diabetic wound healing.…”
