Singlet oxygen scavenging by α‐tocopherol and β‐carotene: Kinetic studies in phospholipid membranes and ethanol solution

Abstract: The rate constants (ks) of 1O2 scavenging for alpha-tocopherol (alpha-Toc) and beta-carotene (beta-Car) were measured in liposome membranes, and compared with those in EtOH solution. 1O2 was site-specifically generated by photoirradiation using two photosensitizers, water-soluble Rose bengal (RB) and lipid-soluble 12-(1-pyrene)-dodecanoic acid (PDA). The ks value for beta-Car in EtOH solution was 1.3 x 10(10) M-1 s-1, which was 36 times that for alpha-Toc (3.6 x 10(8) M-1 s-1), but there was no difference betw… Show more

“…14,15 Its inhibition of peroxidation is generally ascribed to the hydrogen transfer from phenolic hydroxyl groups to the radicals. This is illustrated in Scheme 1, 10, 13 where LOO• and LOOH stand for a lipid peroxyl radical and the corresponding hydroperoxide, respectively.…”

“…We also modified the lipid composition to enhance triggerability. The ROS-sensitive lipid in the EggPC-based liposomes contained singly allylic hydrogens (31) [from methylene groups adjacent to one double bond, which can react with singlet oxygen to form lipid peroxides (32,33)]. A higher reactivity with singlet oxygen can be achieved using lipids with biallylic hydrogens (from methylene groups adjacent to two double bonds) (34).…”

“…Cells treated with pAG became sensitive to UV light exposure as a result of the generation of reactive oxygen species such as singlet oxygen (30). The NRel-4 cell line was more sensitive to UV light killing than the parent strain under the same conditions.…”
