Single intravenous and oral dose pharmacokinetics of florfenicol in the channel catfish (Ictalurus punctatus)

Abstract: Plasma distribution and elimination of florfenicol in channel catfish were investigated after a single dose (10 mg/kg) of intravenous (i.v.) or oral administration in freshwater at a mean water temperature of 25.4 °C. Florfenicol concentrations in plasma were analyzed by means of liquid chromatography with MS/MS detection. After i.v. florfenicol injection, the terminal half-life (t(1/2)), volume of distribution at steady state (V(ss)), and central volume of distribution (V(c)) were 8.25 h, 0.9 and 0.381 L/kg, … Show more

“…In this study, the t 1/2 β in plasma of yellow catfish was 9.63 h with a Cl of 0.18 L/h/kg, which is consistent with the results found in tilapia (10.03 h, Feng and Jia ) and channel catfish (9.11 h, Gaunt et al ). The order of t 1/2 β in different tissues in this study were the following: kidney (15.75 h) = muscle (15.75 h) > skin (14.44 h) > liver (11.55 h) > plasma (9.63 h).…”

“…administration at a dose of 10 mg/kg at 25 C. The mean values of C max and T max in the plasma of yellow catfish were 2.35 µg/mL and 7.25 h, respectively, which were lower and earlier than those in cod (12.40 µg/mL and 9 h after 10 mg/kg of oral doses, Samuelsen et al ), Korean catfish (9.59 µg/mL and 8 h after 10 mg/kg of oral doses, Park et al ), tilapia (4.46 µg/mL and 12 h after 10 mg/kg of oral doses, Feng et al ), and channel catfish, I . punctatus (7.61 ± 0.87 µg/mL and 9.20 ± 2.53 h after after 10 mg/kg of oral doses, Gaunt et al ). The order of the C max values in yellow catfish was the following: kidney > liver > muscle > skin > plasma.…”

“…However, ECVs do define the MIC distribution of WT isolates that are integral, along with pharmacokinetic/pharmacodynamic data and clinical outcome data, to set clinical breakpoints (CLSI 2008(CLSI , 2011. Therefore, our results, in combination with existing pharmacokinetic data (Gaunt et al 2012(Gaunt et al , 2013, data on pharmacodynamics and data on treatment outcomes, could help foster clinical breakpoints for F. columnare.…”
