Single factors alone can induce mesenchymal-like morphology, but not promote full EMT in breast cancer cell lines with different hormone statuses

Abstract: Our results using four different breast cancer cell lines in vitro do not provide evidence that EMT is induced by hypoxia alone or in combination with TGF-β1 or the Notch ligand Jagged-1. The inconsistency in morphological appearance and EMT-markers, as well as the time dependent variation in E-cadherin responses could not support EMT. Importantly, there was not one single common response pattern to the stimuli used, suggesting that cell lines with different hormone statuses display individual traits that resp… Show more

“…In the case of whole-cell analysis, this differentiation disappears. It is a known fact that studied cytoskeletal structures, as well as the cells themselves, are dynamic objects and their behavior and morphology depend on several different factors that are not included in the study [ 57 , 58 , 59 , 60 ]. However, during the experiments, efforts were made to minimize the influence of external factors through identical culturing conditions, passage and staining protocols, or microscopic observation parameters itself.…”

“…In case of microtubular network its area was by 8% lower on glass than in cells grown on PHO substrates (Figure D). The considerable deviations in the results obtained from the analysis (Figure ) resulted from the fact that the studied cytoskeletal structures, as well as the cells themselves, are dynamic objects and their behavior and morphology quantitative analysis was difficult and often burdened with high uncertainties . During the experiments, however, efforts were made to minimize the influence of peripheral factors, that is, breeding method differences or various experimental conditions through identical culturing conditions, passage and staining protocols, and microscopic observation parameters in combination with precise environment control (temperature, humidity, CO 2 level).…”
