Abstract. Recently, a simple method for the extraction of lipids from wet biomass using liquefied dimethyl ether (DME) without drying, cell disruption, or heating was proposed. Here, the versatility of this method was evaluated for labyrinthula Aurantiochytrium limacinum (A. limacinum). The liquefied DME was passed through the extractor that filled by A. limacinum at different time intervals. The extraction of lipids from A. limacinum of moisture-rich microorganism was successfully achieved, the yield of lipid was 46.1 wt% of the dry weight of the sample. In comparison, the yields of lipid were 21.3 wt%, 43.6 wt% and 50.7 wt% when supercritical carbon dioxide (SCCO2), hexane-Soxhlet and Bligh-Dyer (BD) extraction methods were applied as extractants, respectively. However, the drying and cell-disruption process were required in SCCO2, hexane-Soxhlet, and BD extraction methods.

Các acid béo không no (PUFA) có các đặc điểm cấu trúc độc đáo tương ứng với từng chức năng riêng biệt như điều chỉnh sinh lý tế bào và điều chỉnh sự biểu hiện của các gen nhất định. Do đó, khi thiếu hụt PUFA sẽ xuất hiện những bất thường ở da, thận, mạng lưới thần kinh, các phản ứng miễn dịch và viêm, hệ tim mạch, hệ nội tiết, hệ hô hấp và hệ sinh sản. Trong dầu cá, tỷ lệ PUFA tương đối thấp gây khó khăn trong việc sản xuất trên quy mô lớn. Vì vậy, việc thăm dò các nguồn của PUFA đặc biệt là acid arachidonic-AA, acid eicosapentaenoic-EPA, acid docosapentaenoic-DPA/DHA thu hút được nhiều nghiên cứu. Vi tảo biển dị dưỡng (VTBDD) thraustochytrid có khả năng sản xuất lượng lớn DHA và thành phần PUFA đa dạng. DHA có thể được tổng hợp bởi sự chuyển hóa của AA, EPA hay DPA. Các dạng khác nhau của PUFA phản ánh mối quan hệ trong phân loại. Mười vi tảo biển dị dưỡng thraustochytrid phân lập được từ rừng ngập mặn Xuân Thủy, Nam Định chứa thành phần acid béo đa dạng từ C12 -C28, đặc biệt có chứa hai thành phần PUFA quan trọng là EPA và DPA. Tỷ lệ PUFA của mười chủng VTBDD thraustochytrid chiếm 8,95 -49,62% lipid tổng số. DPA so với các PUFA khác đều cao nhất đối với cả mười VTBDD nghiên cứu, chiếm 20,22 -39,35% TFA. Mười VTBDD thraustochytrid sinh trưởng tốt nhất đối với nguồn carbon là glucose, lipid tổng số đạt 7 -12,35% trọng lượng khô sau 72 giờ. Tốc độ sinh trưởng và tổng hợp lipid trên nguồn nitrogen hữu cơ tốt hơn so với nguồn nitrogen vô cơ. Nguồn nitrogen tốt nhất cho sinh trưởng và tổng hợp lipid của VTBDD thraustochytrid là cao nấm men, với lipid tổng số đạt 8,57-18,87% trọng lượng khô sau 72 giờ. Đặc biệt các acid béo không no đa nối đôi (PUFA) như acid docosahexaenoic (DHA), acid docopentaenoic (DPA), acid eicosapentaenoic (EPA) không chỉ hỗ trợ quá trình phát triển chức năng bình thường của hệ thần kinh trung ương mà còn giúp chống lại các căn bệnh nguy hiểm như ung thư, xơ vữa động mạch (Crawford et al., 1997;Li et al., 2014). Abstract:Poly unsaturated fatty acids (PUFAs) participate in activities of all organisms from energy source to structural components. Disruption of metabolism results in diseases like atherosclerosis and obesity. In this work, optimal conditions to production of arachidonic, eicosapentaenoic and gamma-linolenic acids has been conducted in the oil degrading fungi-Cephalosporium humicola IE, Mucor globosus 11 and Pythium irregulare LX isolated from soils in Absheron peninsula, Azerbaijan. The fermentation process carried out at physicochemical conditions, carbon and nitrogen sources, temperature, pH, rate of inoculum, enzyme activity, exogenous fat and phosphate; as well as the stressful situations. Growth in crude oil as optimal carbon source showed that suitable nitrogen was peptone for lipid biosynthesis. In stable temperature the optimal lipid productionoccurred at 28°C, whereas in "temperature shifting technique", greater yields occurred at 28°C for 5 days followed by at 15°C for 2 days. C/N ratio ranged from 4 to 64 and the optimal medium for production was composed of 2.0% glucose and 0.25% yeast extract, with addition of 0.1% KH 2 PO 4 at neutral pH with 3% inoculum. Although the activities of the enzymes varied among the fungi, but the developmental profiles for all enzymes were practically similar. Additive fat and phosphate accelerated growth and enhanced PUFA production. Data showed that the procedure of adaptation of fungi motivated the increase of the rate of membrane phospholipids with a high quantity of PUFAs. This research is considered to be object of PUFA production as food supplements.
