Single-cell gene-expression profiling reveals qualitatively distinct CD8 T cells elicited by different gene-based vaccines

Abstract: CD8 T cells play a key role in mediating protective immunity against selected pathogens after vaccination. Understanding the mechanism of this protection is dependent upon definition of the heterogeneity and complexity of cellular immune responses generated by different vaccines. Here, we identify previously unrecognized subsets of CD8 T cells based upon analysis of gene-expression patterns within single cells and show that they are differentially induced by different vaccines. Three prime-boost vector combina… Show more

“…AFF1 also binds P-TEFb in vivo (25,42), and ENL and AF9 compete for scaffold binding (30). With four, three, and two human homologs of AFF4, ELL, and ENL, respectively, as well as posttranslational modifications of the subunits, including P-TEFb (14,43,44), these data demonstrate the potential for considerable variability in SEC composition. Alternatively, specific complexes may assemble preferentially or cooperatively.…”

“…An analogous approach has been previously and successfully applied to study cell heterogeneity during in vivo CD8 T-cell differentiation (12,13). Recently, single-cell gene expression profiling was shown to allow identification of qualitative differences in CD8 T-cell responses elicited by different gene-based vaccines (14).…”

“…If keeping the 4-th dimension prior to LDA is beneficial for linkage, then keeping the first three axes is also highly relevant, and this is true regardless of the properties of the single-cell trait distributions. We found this very useful: our here, but also for gene expression [43] or parameters describing the micro-environment of the cells [44]. It would therefore be interesting to search for scPTL affecting multiple cellular traits simultaneously instead of treating cellular traits one by one.…”
