Single-Cell Analysis Using Drop-on-Demand Inkjet Printing and Probe Electrospray Ionization Mass Spectrometry

Abstract: This study describes a novel method for single-cell analysis and lipid profiling by combining drop-on-demand inkjet cell printing and probe electrospray ionization mass spectrometry (PESI-MS). Through inkjet sampling of a cell suspension, droplets with single cells were generated, precisely dripped onto a tungsten-made electrospray ionization needle, and immediately sprayed under a high-voltage electric field. Lipid fingerprints of single cells were obtained by a mass spectrometry (MS) detector. A homemade mag… Show more

“…For as-mentioned techniques, analytes are required to be activated with corresponding responses for the analysis of complex mixture [104], MS approach is a more powerful tool, since a wide range of molecules could be label-free, specifically and sensitively detected by it with providing information on the molecular weights and chemical structures of the analytes [105][106][107][108]. Recently, ambient ionization technique has been introduced into MS and draw great attentions.…”

“…By spraying the solvent vapor to the needle tip, metabolites were electrosprayed from the tip of the probe. Recently, Chen et al developed a method for single-cell analysis and lipid pro ling by combining drop-on-demand inkjet cell printing and PESI-MS. 89) rough inkjet sampling of a cell suspension, droplets including single cells with a volume of ∼500 pL were generated. ey were precisely dripped onto a tungsten-made electrospray ionization needle, and immediately spayed under a high-voltage electric eld.…”

“…To better understand and make use of this ionization source, significant efforts have been made over the past decades and have resulted in the developments of a multitude of theories on the mechanism of ionization process, for both direct current (DC) and alternating current (AC) ESI. Based on these theories, a variety of techniques and new ionization sources have also been developed to meet different analytical needs …”
