Simvastatin rescues memory and granule cell maturation through the Wnt/β-catenin signaling pathway in a mouse model of Alzheimer’s disease

Abstract: We previously showed that simvastatin (SV) restored memory in a mouse model of Alzheimer disease (AD) concomitantly with normalization in protein levels of memory-related immediate early genes in hippocampal CA1 neurons. Here, we investigated age-related changes in the hippocampal memory pathway, and whether the beneficial effects of SV could be related to enhanced neurogenesis and signaling in the Wnt/β-catenin pathway. APP mice and wild-type (WT) littermate controls showed comparable number of proliferating … Show more

“…Previous studies have found that Wnt/βcatenin appears to be an important regulator of neurodegenerative diseases including AD. Because of the protective role of Wnt/β-catenin signaling in neurons, activation of Wnt/βcatenin signaling is considered to have potential value in the treatment of neurodegenerative diseases affecting synaptic integrity [64]. Previous studies have reported that β-catenin is downregulated in AD [65,66], which is consistent with our findings in Aβ 1-42 -induced PC12 cells.…”

“…To determine whether the activation of β-catenin is essential for the TBK1-mediated promotion of EMT, XAV939 [ 26 , 27 ] was used to inhibit nuclear entry of β-catenin in TBK1-overexpressing cells. The results showed that inhibition of β-catenin nuclear entry blocked the TBK1-mediated upregulation of vimentin and the downregulation of E-cadherin (Fig.…”

“…Simvastatin is a frequently used statin included in various drug delivery systems and is widely used in the treatment of cardiovascular diseases [45,46], tumors [47] and Alzheimer's disease [48]. Dental implant osseointegration is a specific phenomenon of osteogenesis, and the promotion of osteogenesis can effectively promote dental implant osseointegration.…”
