Simvastatin Inhibits Cardiac Hypertrophy and Angiotensin‐converting Enzyme Activity in Rats With Aortic Stenosis

Abstract: 1. In the present study, we tested the hypothesis that long-term administration of the hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitor simvastatin may regress hypertrophy and the possible effect of simvastatin on angiotensin-converting enzyme (ACE) activity in rats with pressure-overload cardiac hypertrophy. 2. Pressure-overload left ventricular hypertrophy (LVH) of rats was induced by part coarctation of the abdominal aorta; a sham-operated group served as the control. Six weeks after operat… Show more

“…Recent findings that the statins attenuate myocardial hypertrophy in vitro and in vivo provide evidence that these drugs might also be involved in tissue remodeling in the heart [4][5][6]. This view was further supported by three studies showing that statins positively affect left ventricular remodeling in animal models of MI and heart failure [7][8][9].…”

“…Simvastatin significantly reduces hypertrophy in rats with pressure overload due to aortic banding. 98 Likewise, the hypertrophic and cardiomyopathic phenotype of a double-transgenic rat with overexpression of both renin and angiotensinogen is improved by cerivastatin treatment. 99 Fluvastatin increases survival in a murine model of myocardial infarction.…”

“…Statins induce regression of left ventricular hypertrophy in animals and humans with ischemic heart disease (33, 44), an effect that may be mediated via reduction of Ras membrane targeting and activation (45). Although there was a trend for pravastatin treatment to mitigate the increase in LVMI in this study, this was not statistically significant.…”
