Simultaneous quantification of GABAergic 3α,5α/3α,5β neuroactive steroids in human and rat serum

Abstract: The 3α,5α-and 3α,5β-reduced derivatives of progesterone, deoxycorticosterone, dehydroepiandrosterone and testosterone enhance GABAergic neurotransmission and produce inhibitory neurobehavioral and anti-inflammatory effects. Despite substantial information on the progesterone derivative (3α,5α)-3-hydroxypregnan-20-one (3α,5α-THP, allopregnanolone), the physiological significance of the other endogenous GABAergic neuroactive steroids has remained elusive. Here, we describe the validation of a method using gas ch… Show more

“…Higher bioavailability of 5b-AP could contribute to the anticonvulsant effect in vivo despite less overall potentiation of GABAergic current in the slice (Fig. 6A) (Porcu et al, 2009). At elevated concentrations that activate both synaptic and extrasynaptic receptor types, it is feasible that the direct binding of 5b-AP to receptors overcomes the reduced effects on tonic current (Hosie et al, 2006).…”

“…Plasma levels of neurosteroids (ie, pregnenolone, allopregnanolone, pregnanolone (ie, 3α-hydroxy-5β-pregnan-20-one), 3α,5α-androsterone, and 3α,5β-androsterone) were analyzed by gas chromatography with mass spectrometry (GC-MS) at the University of North Carolina at Chapel Hill as described previously (Porcu et al, 2009). Plasma progesterone levels were assayed at the NIH Clinical Center Department of Laboratory Medicine by LC-MS/MS, and luteal plasma levels o2 ng/ml were considered to be consistent with either anovulation or a clinic visit too late or early in the luteal phase and, therefore, were not included in comparisons of plasma neurosteroid levels (see below).…”

“…Ethanol activates the hypothalamic-pituitary-adrenal (HPA) axis (Rivier et al, 1984;VanDoren et al, 2000), increasing 3a,5a-THP and 3a,5a-THDOC in the blood plasma, cerebral cortex, and hippocampus (Barbaccia et al, 1999;Porcu et al, 2009;VanDoren et al, 2000). However, it is not known whether this elevation of GABAergic steroids originates only from the periphery or whether there are changes in levels of GABAergic steroids that occur locally in the brain.…”
