Simultaneous expression by porcine aorta endothelial cells of glycosphingolipids bearing the major epitope for human xenoreactive antibodies (Gal?1?3Gal), blood group H determinant andN-glycolylneuraminic acid

Abstract: Glycosphingolipids were isolated from primary cultures of porcine endothelial cells labelled with 14C-galactose or 14C-glucosamine. They were characterized by their mobility on thin layer chromatogram, their sensitivity to exoglycosidases, and their labelling with antibodies. In addition to the major glycosphingolipids, globotetra- and globotriaosylceramide, minor ones were identified as penta- and heptaglycosylceramide of the neolactoseries terminated by either Gal alpha 1-3Gal- (xenoreactive epitope) or Fuc … Show more

“…Blood group A and H hexaglycosylceramides as well as H pentaglycosylceramides are also abundant constituents (Holgersson et al, 1990b;Bouhours et al, 1997). Human blood group H determinant has also been detected by immunostaining on primary aortic endothelial cells in culture (Bouhours et al, 1996). These findings are in contrast to a study in which immunofluorescent staining of different pig tissue failed to detect blood group determinants in blood vessels .…”

“…This sulfate and glucuronic acid carrying GSL was also reported to be present in rat brain microvessels by the same group (Miyatani et al, 1990), indicating prevalence for brain vascular cells of different species. Primary cultures of porcine endothelial cells revealed major neutral GSLs of the globo-series (Bouhours et al, 1996), thus showing some homology to endothelial cells of human origin (Gillard et al, 1987). However, the overall feature of all endothelial cells analyzed so far lies in the dominance of G M3 .…”

“…In a previous analysis of sera from the patients transplanted with porcine ICCs, Abs with non-Gal reactivity were detected in some of them (26) (34)(35)(36), has been widely discussed as a potential non-Gal antigen (13,37 (28). The Neu5Aca3Galb3GlcNAcb-structure, also known as the DU-PAN-2 antigen (41,42),…”
