Simultaneous determination of three flavonoids and one coumarin by LC–MS/MS: Application to a comparative pharmacokinetic study in normal and arthritic rats after oral administration of Daphne genkwa extract

Abstract: A selective and sensitive liquid chromatography-tandem mass spectrometry method was developed and validated for investigating the pharmacokinetics of umbelliferone, apigenin, genkwanin and hydroxygenkwanin after oral administration of Daphne genkwa extract. Plasma samples were treated by protein precipitation with acetonitrile. Analytes were detected by triple-quadrupole MS/MS with an ESI source in negative selection reaction monitoring mode. The transitions of m/z 161 → 133 for umbelliferone, m/z 269 → 117 fo… Show more

“…This peak showed a parent ion at m/z 161 [M-H]ion and characteristics MS/MS fragments at m/z 133 due to loss of CO moiety, m/z 117 corresponding to the loss of CO 2 and m/z 105 produced as a result of loss of 2CO. The proposed mass fragments resulting from the fragmentation of 7-Hydroxy-2Hchromen-2-one is shown in (Figure 2) and is in agreement with previously published result (Zhou et al, 2018).…”

“…For instance, after oral administration of radiolabeled apigenin or the flower extract of Daphne genkwa , a plasma concentration peak was obtained after 1 h and reached approximately 40–50 ng/mL in rats. Moreover, these studies showed the sustained persistence of apigenin, suggesting an accumulation in tissues [11,12]. However, in humans, after consumption of parsley or chamomile extract, the plasma concentration of apigenin remains low (<1 µM), possibly due to its lower bioavailability in humans than in rodents and different metabolism by the intestinal microbiota [9,13].…”

“…Similarly, 29 [M-H] − at m / z 161.023 gave a base peak at m / z 133.028, due to the loss of CO group, and fragment ions at m / z 115.018 [M-H-CO-H 2 O] − and m / z 105.033 [M-H-2CO] − . 29 and 27 differ from 31 by one and two OH groups and could be related to umbelliferone and aesculetin, respectively ( Table 2 ) [ 21 ]. In the fragmentation pattern of 30 and 32, the base peaks at m / z 192.005 [M-H-CH 3 • ] − and 176.011 [M-H-CH 3 • ] − corresponded to the loss of CH 3 • radical.…”
