A highly sensitive and simple spectrofluorimetric method was developed for the determination of loratadine (LRT) and desloratadine (DSL) in their pharmaceutical formulations. The proposed method is based on investigation of the fluorescence spectral behaviour of LRT and DSL in a sodium dodecyl sulphate (SDS) micellar system. In aqueous solution of acetate buffer of pH 4.5, the fluorescence intensities of both LRT and DSL were greatly enhanced (240%) in the presence of SDS. The fluorescence intensity was measured at 438 nm after excitation at 290 nm for both drugs. The fluorescence-concentration plots were rectilinear over the range 0.05-2.0 µg/mL for both LRT and DSL, with lower detection limits of 5.13 × 10(-3) and 6.35 × 10(-3) µg/mL for LRT and DSL, respectively. The method was successfully applied to the analysis of the two drugs in their commercial tablets, capsules and syrups, and the results were in good agreement with those obtained with the official or comparison methods. The proposed method is specific for the determination of LRT in the presence of other co-formulated drugs, such as pseudoephedrine. The application of the proposed method was extended to stability studies of LRT and DSL after exposure to different forced degradation conditions, such as acidic, alkaline and oxidative conditions, according to ICH guidelines.

Two simple, sensitive and inexpensive spectrophotometric methods have been described for the determination of loratadine, an antiallergic drug. The procedure was based on the formation of coloured chargetransfer complexes of loratadine acting as an n-electron donor with iodine acting as σ-acceptor in chloroform (method A) and 2,3,5,6-tetrachloro-1,4-benzoquinone (p-chloranil) acting as π-acceptor in dimethyl sulfoxide (method B). The optimizations of various experimental conditions and the mole ratio were studied. The chargetransfer complexes formed were found to absorb at 399 nm and 528 nm. Beer's law was obeyed for these methods in a concentration range of 6.5-78 mM and 0.2-6 µg ml -1 with correlation coefficients 0.9999 and 0.9993. Application of these methods to the assay of loratadine in tablet form was described and the results were statistically validated. The results of analysis of commercial formulations and the recovery study of the drug suggested that there was no interference from any excipients, which were present in pharmaceutical formulations.

A new highly sensitive and specific spectrofluorimetric method has been developed to determine a sympathomimetic drug pseudoephedrine hydrochloride. The present method was based on derivatization with 4-chloro-7-nitrobenzofurazan in phosphate buffer at pH 7.8 to produce a highly fluorescent product which was measured at 532 nm (excitation at 475 nm). Under the optimized conditions a linear relationship and good correlation was found between the fluorescence intensity and pseudoephedrine hydrochloride concentration in the range of 0.5-5 µg mL(-1). The proposed method was successfully applied to the assay of pseudoephedrine hydrochloride in commercial pharmaceutical formulations with good accuracy and precision and without interferences from common additives. Statistical comparison of the results with a well-established method showed excellent agreement and proved that there was no significant difference in the accuracy and precision. The stoichiometry of the reaction was determined and the reaction pathway was postulated.
