1. The possible involvent of noradrenaline, dopamine, serotonin and 5-hydorxyindoleacetic acid in the behavioural reinforcement produced by invravenous cocaine has been investigated. 2. rats, in Skinner boxes, were allowed to self-administer cocaine through a chronically implanted jugular vein cannula. A consistent response rate in the order of one self-administration every 10 min was observed with a dose of 3.0 mg/kg cocaine hydrochloride per response. 3. Another group of rats was cannulated and injected with a range of one to six such doses of cocaine at 10 min intervals on the basis of the responding rate, established by rats in the Skinner box. At times of 2,5 and 8 min after each dose of cocaine, the rats were decapitated and brain concentrations of noradrenaline, dopamine, serotonin and 5-hydroxyindoleacetic acid were determined. 4. The brain levels of noradernaline exhibited a cyclic pattern of rise and fall after each dose of cocaine, which corresponded in its time base with the time interval between responses established by the rats in the Skinner box.

“…The hypothalamus was dissected free immediately after decapitation and consisted of the tissue 2 mm on each side of the midline extending from the rostral edge of the optic chiasma to the base of the mammillary bodies and dorsally to the level of the anterior commissure. The tissue was immediately weighed and frozen on dry ice, stored at -20 °C and assayed for either catechol amines [2,3] or indolamines [4,5] by spectrofluorophotometric methods following elution of the homogenate from a weak cationexchange column (Bio-Rex 70). Data were expressed as micro grams of monoamine per gram tissue wet weight and analyzed stat istically with analysis of variance and with the Student-NewmanKeuls test for multiple comparisons.…”

Diurnal Variation in Hypothalamic Monoamines and Plasma Corticosterone in the Turkey after Inhibition of Tyrosine Hydroxylase or Tryptophan Hydroxylase

Circadian rhythms occur in hypothalamic serotonin content in the turkey. Inhibition of serotonin (5-HT) synthesis by parachlorophenylalanine (PCPA) blocks circadian variation in hypothalamic serotonin levels suggesting that the major cause of diurnal fluctuations is differing rates of synthesis of the amine. PCPA treatment also abolishes the diurnal rhythm in plasma corticosterone and elevates the basal levels of this steroid suggesting that 5-HT terminals in the hypothalamus may inhibit the release of ACTH at certain times of the day and in this fashion generate diurnal fluctuations in plasma corticosterone. Hypothalamic levels of dopamine (DA) and norepinephrine (NE) also vary diurnally, and synthesis inhibition with alpha-methyl-para-tyrosine abolishes the diurnal rhythm in NE but not DA or in plasma corticosterone. Temporal changes in release and/or degradation of DA are significant factors contributing to the diurnal rhythms of this amine in the hypothalamus. Diurnal fluctuations in plasma corticosterone do not depend on recent synthesis of catecholamines.

1 The effect of pretreatment with clomipramine hydrochloride (15 mg/kg, i.p.) on the (-)-trans-A9-tetrahydrocannabinol (A9-THC)-induced changes in body temperature and brain amines of the rat was investigated. 2 A dose of 0.05 mg/kg of A9-THC produced hyperthermia and a decrease in whole brain concentration of 5-hydroxyindoleacetic acid (5-HIAA). Doses of 2 and 5 mg/kg produced hypothermia and increases in brain 5-HIAA whereas 0.5 mg/kg did not affect either parameter. A9-THC, at any of the doses, did not affect the whole brain concentrations of dopamine, noradrenaline or 5-hydroxytryptamine. 3 Clomipramine modified these responses of A9-THC in that the dose-response curves appeared to be shifted to the right. 4 It is concluded that clomipramine acts as an antagonist to these actions of A9-THC by interfering with entry of A9-THC into tryptaminergic neurones.
