Simultaneous Determination of Chlorophylls, Pheophytins, β‐Carotene, Tocopherols, and Tocotrienols in Olive and Soybean Oils by High‐performance Liquid Chromatography

Abstract: A high-performance liquid chromatographic method is described for the simultaneous measurement of chlorophylls, pheophytins, ␤ ␤ ␤ ␤ ␤-carotene, tocopherols, and tocotrienols in vegetable oils using 2 spectrophotometers in series and 1 normal-phase silica column. Oil samples were diluted in the mobile phase, isopropanol-hexane (1.5:98.5 vol/vol), and injected directly onto the column. A programmable UV/Vis spectrophotometer was used to measure chlorophylls a and b, pheophytins a and b, and ␤ ␤ ␤ ␤ ␤-carotene a… Show more

“…In some cases, the direct solvent extraction (Mendoza, Pons, Bargallo, & Lopez-Sabater, 2003;Ryynänen, Lampi, Salo-Väänänen, Ollilainen, & Piironen, 2004) or the Soxhlet extraction (González, Pablos, Martín, León-Camacho, & Valdenebro, 2001;Ramadan & Moersel, 2002) of the vitamins, without previous saponification, is carried out. For the analysis of vitamin E in oils, the sample can be injected directly into the LC system after appropriate dilution in an organic solvent (Gliszczynska-Swiglo & Sikorska, 2004;Puspitasari-Nienaber, Ferruzzi, & Schwartz, 2002;Seppanen, Rahmani, & Csallany, 2003).…”

“…For all these reasons, it is not surprising that a great effort is dedicated to develop standardized methods to quantify pigments and possibly correlating them to specific features of EVOO production and quality. Several analytical methods based on high performance liquid chromatography (HPLC) have been proposed to identify and quantify carotenoids (Cortes et al, 2004), chlorophyll derivatives (Watanabe, 1984) or both (Seppanen, Rahmani, & Csallany, 2003;Minguez-Mosquera, Gandul-Rojas, & Gallardo-Guerrero, 1992). Most of these methods require a preventive solvent extraction of pigments from the olive oil lipid matrix.…”
