Simultaneous Determination of Abscisic Acid and Its Catabolites by Hydrophilic Solid-Phase Extraction Combined with Ultra High Performance Liquid Chromatography–Tandem Mass Spectrometry

Abstract: An efficient and selective pretreatment method of one-step hydrophilic interaction chromatography-based solid phase extraction (HILIC SPE) was developed using silica as the sorbent to quickly and sensitively detect endogenous ABA and its five catabolites in fresh Oryza sativa tissues. The extracted analytes were sensitively quantified with ultra high performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS). Under the optimized conditions, good linearity of the developed analytical method was ob… Show more

“…The extraction and determination of ABA, phaseic acid (PA), and dihydrophaseic acid (DPA) were performed by following the protocol described in our previous reports. 41,44 Briefly, [ 2 H 6 ] ABA (Olomouc), [ 2 H 3 ] PA (National Research Council Plant Biotechnology Institute), and [ 2 H 3 ] DPA (National Research Council Plant Biotechnology Institute) were used as internal standards. Plant tissues of 50 mg were frozen in liquid nitrogen and ground into fine powder.…”

“…Several extraction methods such as SPE, dispersive solid-phase extraction (DSPE), and matrix solid-phase dispersion (MSPD) have been developed [ 18 , 19 ]. SPE affords high recovery and remarkable reproducibility, but the adsorbent amount required is large (tens to hundreds of milligrams), and it often requires an activation step (pretreatment with methanol and water) before loading the sample solution [ 20 , 21 ]. In DSPE, the adsorbent is uniformly dispersed to ensure sufficient contact with the sample solution, washing solvent, and elution solvent, such that adsorption and desorption occur rapidly [ 22 , 23 ].…”

“…Among many techniques, solid-phase extraction (SPE) is considered one of the most effective techniques for coextraction due to its convenience, robustness, and rapidity. Therefore, SPE was used here for the analysis of tea samples [14,15].…”
