Objective: The prevalence of Hypertension in India is estimated to be greater than 45% in the middle age population; elevated hypertension is one of the known causes of CHD, strokes and death. The objective of this work was to develop a simple, accurate and robust HPTLC method was developed for simultaneous estimation of Valsartan and Hydrochlorothiazide in bulk and tablets. Methods: The mobile phase optimized by HPTLC method consists of Toluene: Ethyl acetate: formic acid in the ratio of (3:7:0.3, v/v/v). The solvent front was a run-up to a distance of 80 cm; which took 15 min for the development of TLC plate. Analytical Wavelength for UV detection selected was at 225 nm. Results: Retention factor was found to be 0.74±0.02 and 0.45±0.02 for VAL and HTZ, respectively. Developed chromatographic method was validated as per ICH, Q2(R1) guidelines. Linearity was found at concentration range 300-1800 ng/spot (r2 = 0.999), 50-300ng/spot (r2 = 0.9991) for VAL and HTZ respectively. LOD were found to be 23.02 ng/spot and 4.12ng/spot respectively for Valsartan and Hydrochlorothiazide. LOQ was found to be 69.76ng/spot and 12.5ng/spot for, Valsartan and Hydrochlorothiazide respectively. Conclusion: Percentage RSD was found out to less than 2, indicating the developed method was precise and can be successively applied to pharmaceutical formulation. No interferences with the excipients were reported.

FDA has recently approved a new fixed-dose combination of amlodipine besylate (AMD) and celecoxib (COX) for the treatment of hypertension and osteoarthritis. No analytical method has been reported for analysis of these two analytes so far. Hence, to monitor the quality and quantity in the formulation of AMD and COX a simple, accurate, precise, economical, and eco-friendly spectroscopic analytical method has been established. The first method involves the determination of AMD and COX by the first derivative UV spectroscopic method with scaling factor 10. However, the second method was based on the direct measurement of UV absorbance of AMD at 364.3 nm and ratio first derivative UV spectroscopic method for COX. Both methods showed good linearity in the range of 5 to 40 μg/ml for COX, whereas AMD showed linearity in the range of 0.5 to 10 μg/ml in first derivative method with scaling factor 10 and 1 to 10 μg/ml in the second method with good correlation coefficient (R2 > 0.998). Both the methods were validated for LOD, LOQ, accuracy, precision, recovery studies, and stability as per the ICH guidelines, and the validated results were well within the acceptable range. Both the methods were successfully utilized for the determination of AMD and COX in the presence of each other in the formulation, and statistically compared between the proposed methods. Therefore, the proposed procedures can be utilized for regular quality control studies.

Two new, simple, fast and sensitive spectrophotometric methods were developed for analysis of binary mixtures of Lisinopril (LIS) and hydrochlorothiazide (HCT). These methods were absorptivity factor method and ratio subtraction method. In the first method, calculations were made using the absorbance of the mixtures at 211 nm where LIS absorptivity is half HCT absorptivity. On the other hand, the second method was based on obtaining the original spectrum of LIS from the zero order spectrum of the binary mixture then determining the concentration of LIS from its linear regression equation. HCT concentration in both methods was determined from the absorbance at 270 nm where a lack of interference of LIS occurs. The methods were validated according to ICH and applied successfully on different ratios of laboratory mixtures and on pharmaceutical tablets with excellent accuracy and precision.
