Simultaneous CRISPR/Cas9-mediated editing of cassavaeIF4EisoformsnCBP-1andnCBP-2reduces cassava brown streak disease symptom severity and incidence

Abstract: SummaryCassava brown streak disease (CBSD) is a major constraint on cassava yields in East and Central Africa and threatens production in West Africa. CBSD is caused by two species of positive sense RNA viruses belonging to the family Potiviridae, genus Ipomovirus: Cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV). Diseases caused by the family Potyviridae require the interaction of viral genome-linked protein (VPg) and host eukaryotic translation initiation factor 4E (eIF4E) iso… Show more

“…In cassava, there are five genes encoding eIF4E proteins, of which only nCBP-1 (novel cap-binding protein-1) and nCBP-2 associate with VPgs (viral genome-linked proteins). Consistently, only the ncbp-1/ncbp-2 double mutants generated through CRISPR/Cas9 showed delayed and attenuated symptoms after infection with CBSV (Cassava brown streak virus) [60].…”

“…In the third place, the process of breeding numerous woody horticulture crops like apple and walnut can take as long as twenty to thirty years to produce a single individual that possesses a combination of several desirable characteristics. Even while molecular markerassisted selection and fast track breeding techniques have the potential to speed up the breeding and selection processes, this still takes a significant amount of labour and land resources of an enormous magnitude [49]. During the course of crop evolution, there were instances in which spontaneous variations with novel features emerged, and these types of variations have been maintained.…”
