Simultaneous Application of Photothermal Therapy and an Anti‐inflammatory Prodrug using Pyrene–Aspirin‐Loaded Gold Nanorod Graphitic Nanocapsules

Abstract: Photothermal therapy( PTT) has been extensively developed as an effective approach against cancer.H owever, PTT can trigger inflammatory responses,i nt urn simulating tumor regeneration and hindering subsequent therapy. A therapeutic strategy was developed to deliver enhanced PTT and simultaneously inhibit PTT-induced inflammatory response.1 -Pyrene methanol was utilizet os ynthesize the anti-inflammatory prodrug pyrene-aspirin (P-aspirin) with ac leavable ester bond and also facilitate loading the prodrug on … Show more

“…We chose to supplement PTT because the heat generation in the tumor tissue could naturally create an acute inflammatory environment for the recruitment of neutrophils with our NPNs from the blood circulation 26 . In addition, PTT is known to suffer from tumor recurrence due to the incomplete eradication of microtumors by the shallow light penetration depth in the tumor tissue 27 . It is therefore of great importance to develop combinatorial therapeutic methods to supplement the PTT efficacy.…”

“…Moreover, aspirin synergistically sensitizes osteosarcoma cells to cisplatin and radiotherapy . Combination of photo‐thermal therapy and aspirin has promising anticancer effects . Randomized clinical trials are necessary to evaluate the efficiency of combined cancer therapy including aspirin, molecular targeted therapy, chemotherapy, and radiotherapy.…”

“…Notably, the aspirin‐like molecule o‐(acetoxyphenyl)hept‐2‐ynyl sulfide inhibits COX2 much more potently and selectively than aspirin . In addition to the above‐described derivatives of aspirin, an alternative strategy is to incorporate aspirin into nanoparticles . It warrants further studies to test the efficacy of these novel aspirin derivatives in clinical setting.…”

“…Based on this strategy, anti-in ammatory prodrug pyrene-aspirin loaded on gold nanorod-encapsulated graphitic nanocapsule was constructed to achieve photothermal ablation of tumor and simultaneously alleviate PTT-triggered in ammation. [12] This photothermal-antiin ammatory strategy overcomes the key shortcoming of traditional PTT (the trigger of proin ammatory cytokines during PTT). Alternatively, aiming at weakening the in ammatory response during PTT, a CO nanogenerator was constructed with partially oxidized in disul de nanosheets, tumor targeting polymer (PEG-cRGD), and chemotherapeutic drug doxorubicin.…”
