Simultaneous and Sensitive Measurement of Anabasine, Nicotine, and Nicotine Metabolites in Human Urine by Liquid Chromatography–Tandem Mass Spectrometry

Abstract: Methods:We developed a sensitive method for analysis of nicotine and five major nicotine metabolites, including cotinine, trans-3-hydroxycotinine, nicotine-N-oxide, cotinine-N-oxide, and nornicotine, in human urine by liquid chromatography coupled with a TSQ Quantum triple quadrupole tandem mass spectrometer (LC/ MS/MS). Urine samples to which deuterium-labeled internal standards had been added were extracted with a simple solid-phase extraction procedure. Anabasine, a minor tobacco alkaloid, was also included… Show more

“…Tuomi et al (29) and Xu et al (30) described LC-MS/MS methods involving an off-line (manual) SPE cleanup for nicotine and cotinine and had detection limits of 1 to 10 ng/mL and 0.1 to 1 ng/mL, respectively. Similarly, the method described by Heavner et al (31) involved also an off-line SPE and had detection limits of 4.4 and 3.7 ng/mL for nicotine and cotinine, respectively.…”

“…12,16,18 The measurement of cotinine is not an urgent investigation. In our centre, we run the cotinine assay weekly to assess the smoking status of patients who are being considered for cardiothoracic transplantation while attending the assessment clinics.…”

“…A number of reports [35][36][37][38][39][40][41] on determination of nicotine and related alkaloids by LC-MS/MS in different matrices (especially in biological fluids, like urine, plasma, saliva) have been published. However, these methods can be also suitable for e-liquids and aerosol from ECs, but their applicability to these matrices has to be verified.…”
