In Japan, crude drugs that are used as ingredients of kampo formulae are mainly imported from China. In China, cypermethrin and fenvalerate, which are pyrethroid pesticides, are detected frequently in food exported to Japan. In fact, cypermethrin and fenvalerate have been detected in crude drugs distributed in Japan. There is concern that cypermethrin and fenvalerate will move from crude drugs to kampo products. Cypermethrin and fenvalerate in kampo products are controlled by a self-imposed residual pesticide limit set by the Japan Kampo Medicine Manufacturers Association. Several methods to analyze cypermethrin and fenvalerate in kampo products have been reported, but they are time-consuming, require expensive column clean-up, and use large amounts of organic solvents. In this study, a simple, rapid, and inexpensive sample preparation method was developed to determine cypermethrin and fenvalerate levels in kampo products by gas chromatography (GC)-mass spectrometry (MS) with negative chemical ionization (NCI). Twenty-two samples of kampo products were analyzed according to the proposed method. No samples were contaminated by cypermethrin and fenvalerate levels greater than the detection limit.

In Japan, maximum residue levels (MRL) have been set for eight pesticides (a-BHC, b-BHC, g-BHC, d-BHC (BHCs), p,p′ -DDE, o,p′ -DDT, p,p′ -DDD, p,p′ -DDT (DDTs)) in 14 crude drugs (below 0.2 ppm as total of BHCs, below 0.2 ppm as total of DDTs). There are fears that pesticides present in crude drugs for which MRL are set will be changed from BHCs and DDTs to other pesticides with MRL setting as the turning point. There are few surveys of pyrethroid pesticide in crude drugs distributed in Japan. The actual situation of pyrethroid pesticides in crude drugs distributed in Japan after setting MRL is not unclear and should be clariˆed. Although a method to analyze permethrin, cypermethrin and fenvalerate in 11 crude drugs was reported, it is not adequate because the recovery rates of permethrin, cypermethrin and fenvalerate from Cinnamomi cortex were very low and the method, including liquid-liquid partition is di‹cult. In this study, we developed a method using solid-phase extraction to analyze permethrin, cypermethrin and fenvalerate in Cinnamomi cortex with acceptable recovery rates. The sample solution was determined by gas chromatography/mass spectrometry with negative chemical ionization. The recovery rates of permethrin, cypermethrin and fenvalerate from Cinnamomi cortex were between 87.9 and 90.7％. Five samples of Cinnamomi cortex were analyzed according to the proposed method. No samples contained permethrin, cypermethrin and fenvalerate over detection limits. The proposed method could analyze permethrin, cypermethrin and fenvalerate in all crude drugs for which MRL are set.

The search on endocrine disrupting chemicals (EDCs) in non-fatty food was evaluated. A fast, high-throughput, accurate, multiresidue method for the analysis of selected EDC pesticides in fruit and vegetable food samples was developed. The QuEChERS technique was used for sample preparation. Fast GC-MS was performed with a narrow-bore capillary column and a quadrupole benchtop detector with electron ionization (EI) and negative chemical ionization (NCI). A part of the work was devoted to the comparison of NCI versus EI approach concerning the sensitivity of detection and to the study of selectivity enhancement in NCI mode. Matrix-matched standard solutions were utilized for calibration. The methods validation was performed. Fortification studies at 1, 5, 10 and 250 mg kg À1 for 35 pesticides in EI mode and 0.1, 1, 5 and 250 mg kg À1 for 28 pesticides in NCI mode were performed. Average recoveries for each fortification level ranged from 70 to 110% with 480% of recoveries between 90 and 110%. Limits of quantification (LOQs) were established at 5 mg kg À1 for EI and at 1 mg kg À1 for NCI mode, which is lower than the lowest maximum residue level (MRL) value set by the European Commission in fruit and vegetables. The developed and validated fast GC-MS method was successfully applied to the search of EDC pesticides at ultratrace concentration level in real fruit and vegetable samples in Slovakia. Thirty-four samples of 20 different commodities were analyzed. Seven samples contained residues of three or more EDCs pesticides.
