Simulations of Inositol Phosphate Metabolism and Its Interaction with InsP3-Mediated Calcium Release

Abstract: Inositol phosphates function as second messengers for a variety of extracellular signals. Ins(1,4,5)P(3) generated by phospholipase C-mediated hydrolysis of phosphatidylinositol bisphosphate, triggers numerous cellular processes by regulating calcium release from internal stores. The Ins(1,4,5)P(3) signal is coupled to a complex metabolic cascade involving a series of phosphatases and kinases. These enzymes generate a range of inositol phosphate derivatives, many of which have signaling roles of their own. We … Show more

“…5c-d). Notwithstanding, the Michaelis-Menten constant of IP 3 -3K for its substrate is experimentally reported to be K 3 ≈ 1 μM [56,59] and it is likely that intracellular IP 3 levels can reach such micromolar concentrations in vivo [73]. Therefore, in the following, we will keep the Michaelis-Menten formulation for v 3K (12).…”

“…3). Furthermore, one might argue that the inherent inadequacies of "IP 7 " as a nomenclature have led it occasionally-and incorrectly-to be defined as either a "heptaphosphate" (Mishra and Bhalla, 2002) (i.e., an entirely mythical beast endowed with a chain of seven phosphates) or a "heptakisphosphate" (Lee et al, 2007) (which would require seven phosphates to each be individually attached to a carbon-impossible, of course, because inositol has only six carbons). The usage of "IP 7 /IP 8 " should also be avoided because it can obscure the fact that InsP 6 is not the only precursor for this class of compounds; PP-InsP 4 is a member of this family, but it is synthesized from InsP 5 (Fig.…”

“…This model consists of about 100 non-linear ordinary differential equations (ODEs) and algebraic equations and about 200 parameters. Similarly, a detailed model for calcium signalling consists of about 200 equations and even higher number of parameters [9]. The complexity becomes even more appreciable when a network model corresponding to the whole cell, possibly resulting in tens of thousands of non-linear mixed (both continuous and discrete variables) equations with a similar number of parameters, needs to be studied.…”
