Simple Noninvasive Measurement of Skin Autofluorescence

Abstract: Accumulation of advanced glycation end products (AGEs) is thought to play a role in the pathogenesis of chronic complications of diabetes mellitus and renal failure. Several studies indicate that AGE accumulation in tissue may reflect the cumulative effect of hyperglycemia and oxidative stress over many years. Simple quantitation of AGE accumulation in tissue could provide a tool for assessing the risk of long-term complications. Because several AGEs exhibit autofluorescence, we developed a noninvasive autoflu… Show more

“…Although the reference standard for measuring AGE levels in tissues is biochemical determination in skin biopsies, skin autofluorescence (SAF) has emerged as a noninvasive, accurate, and practical method for the analysis of AGE levels in skin 11, 12. SAF‐indicated AGE levels are elevated in diabetes mellitus and end stage renal disease, and some studies suggest that they are associated with CVD independent of known CVD risk factors 13, 14, 15.…”

“…[11] A black light tube, with a peak wavelength of 370 nm is used to illuminate a small region of the skin on the forearm. An optical fiber detects the emission and reflected excitation light, and a spectrometer is used to measure the intensity spectrum.…”

“…Recently, a non-invasive method to measure the skin's content of AGEs using their autofl ourescent characteristic, the AGE-Reader (DiagnOptics B.V., Groningen, The Netherlands), has been developed (107)(108)(109). The in vivo measurements of fl uorescent AGEs correlated strongly with those taken from skin biopsies specimens, as well as with the non-fl uorescent forms of AGEs (110)(111)(112).…”
