Simple hormonal regulation of somatic embryogenesis and/or shoot organogenesis in caryopsis cultures of Pogonatherum paniceum (Poaceae)

Abstract: Pogonatherum paniceum (Poaceae) is a perennial plant with good potential for eco-recovery and ornamental function. This study presents in vitro culture systems of simple hormonal regulation of somatic embryogenesis and shoot organogenesis from mature caryopses. Mature caryopses of P. paniceum were grown on Murashige and Skoog medium with 3% sucrose (w/v) and various concentrations or combinations of 2,4-dichlorophenoxyacetic acid (2,4-D), a-naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BAP). Morphologi… Show more

“…High auxin concentrations rapidly change the metabolism of cells resulting in the development of callus (Dudits et al 1995). The auxin 2,4-D alone or in combination with other plant growth regulators has been widely used for the induction of somatic embryogenesis in many grasses such as Cenchrus ciliares (Colomba et al 2006), Chloris gayana (Gondo et al 2007), Cynodon dactylon (Zhang et al 2007), Zoysia matrella (Dhandapani et al 2008), Eremochloa ophiuroids (Barampuram et al 2009), Pogonatherum paniceum (Wang et al 2008), and U. brizantha (Cabral et al 2011). However, other synthetic auxins such as picloram have not been tested for regeneration of tropical grasses from the Urochloa genus.…”

“…Generally, auxins are used for callus induction and proliferation, and both cytokinins and auxins are required for redifferentiation of callus into organized cells (Wang et al 2008). In this present study, callus was successfully induced in the presence of auxin (IAA) and cytokinin (Zn, Kin).…”

“…Generally, auxins are used for callus induction and proliferation, and both cytokinins and auxins are required for redifferentiation of callus into organized cell (Wang et al 2008). In this study, successful induction of callus was achieved in the presence of NAA, and callus was then subsequently induced to undergo somatic embryogenesis in the presence of BA and NAA.…”
