FOREWORDWhereas the discussion of epitope-antigen structure relating to B cells would not be very different from a similar consideration a decade ago, the nature of T-cell recognition and activation by "antigen" encompasses a completely altered area of concern. Within the past 10 years, concepts of restriction to major histocompatibility-complex MHC molecules and in volvement of T cells in idiotypic interactions have revolutionized the view of the "antigen" that activates the T cells.In this article we use a special definition for the term antigen structure, which includes all elements shown to be recognized by the diversity ofT-cell subpopulations: epitopes (from the nominal antigen), idiotopes (on secreted Ig or on other cell surfaces), and at least two types of MHC determinants, which will be defi ned later.We do not undertake a review of the basis and evidence for MHC restric tion, or T-cell idiotypic interactions: earlier excellent reviews on the former (57,139,148,186) and latter (47,82,83,175) exist, together with several in the present volume. Our focus is confined to current information describ ing the complexity of structures involved in T-cell activation.

Idiotypic Lymphocytes in the Rabbit: Occurrence and Nature of Idiotypic Lymphocytes in Normal and Hyperimmunized Rabbits

Rabbits were hyperimmunized with Micrococcus Pysodeicticus, leading to homogeneous antibody responses. Peripheral blood lymphocytes were taken from the rabbits before and monthly (during 3 months) after the start of the immunization. The cells were stored frozen. Lymphocytes were tested with anti-idiotypic conjugates for the presence of surface idiotypic structures. The nature of the idiotype-positive cells was determined with respect to the presence of IgM or T-cell antigenic determinants on their surface. A sharp rise and fall in the percentage of idiotypic lymphocytes was found, ranging between 1/40,000 and 1/1,000. Initially almost all idiotypic lymphocytes were IgM-positive. In the blood taken 2 months after the start of the immunization 20% of the idiotypic cells belonged to the T-cell population and 10% were negative for both IgM and T-cell antigenic determinants.

A xenogeneic antiserum raised to antireovirus immunoglobulin was used to define an idiotypic determinant present on antibodies to reovirus type 3 hemagglutinin. The same idiotype was identified on nonimmune lymphoid cells and on neuronal cells that specifically bind the hemagglutinin of type 3 reovirus. This idiotypic determinant, called Id3, is shared by (a) a monoclonal antibody to the neutralization site of hemagglutinin from type 3 reovirus; (b) BALB/c serum antibodies to the hemagglutinin of reovirus type 3; (c) R1.1, a murine thymoma cell line that binds reovirus type 3; (d) primary cultures of murine neuronal cells. The presence of an idiotype shared by antihemagglutinin antibodies and by structures on nonlymphoid cells suggests a general relationship between disparate receptors that recognize a common determinant. Furthermore, this suggests a novel approach for the study of viral receptor interactions and for analysis of mechanisms of autoimmune responses.
