Silver nanoparticles offer a synergistic effect with fluconazole against fluconazole-resistant Candida albicans by abrogating drug efflux pumps and increasing endogenous ROS

Abstract: Objectives: A frequent emergence of drug resistance has been observed and posed great threat to global public health recently. This work aimed to investigate the potential synergistic effect and the underlying mechanisms of AgNPs-uconazole combination more extensively through 2 clinically isolated uconazole-resistant Candida albicans (C. albicans) strains. Methods: Antifungal properties of AgNPs and uconazole alone or together against planktonic cells and bio lms were tested. Cellular and molecular targets asso… Show more

“…Furthermore, Ag-NPs are promising therapeutic agents against pathogenic microbes and malignant cells [ 31 , 32 ]. There are numerous reports of Ag-NPs acting as potent biocidal agents against Gram-positive and -negative bacteria and various pathogenic fungi such as Candida albicans [ 33 , 34 , 35 ]. Moreover, Ag-NPs showed high therapeutic activity and selectivity against different malignant cells such as MCF-7, HepG2, Caco-2, HCT-116, and others [ 15 , 36 , 37 ].…”

“…Due to their modest size-to-volume ratio and high thermal stability, metal nanoparticles are widely used in biological domains [ 12 ]. To date, many metal-based nanoparticles have been synthesized by different physical and chemical methods and have been found to have antibacterial properties [ 13 ]. Silver nanoparticles are the best among the various forms of metallic nanoparticles because of their broad antibacterial properties [ 14 , 15 ].…”

“…Yet, in order to limit the amount of silver administered to the body, it is usually used with a substance that presents a synergistic effect. Thus, silver is being used with norfloxacin or streptomycin to enhance the antibacterial effect or together with fluconazole to enhance the antifungal properties [ 51 , 52 ]. AgNPs can also be combined with substances that present immunomodulatory effects and accelerate the overall wound healing process [ 53 ].…”
