Silk derived formulations for accelerated wound healing in diabetic mice

Abstract: Background The present study aimed to prepare effective silk derived formulations in combination with plant extract (Aloe vera gel) to speed up the wound healing process in diabetic mice. Methods Diabetes was induced in albino mice by using alloxan monohydrate. After successful induction of diabetes in mice, excision wounds were created via biopsy puncture (6 mm). Wound healing effect of silk sericin (5%) and silk fibroin (5%) individually and in combination with 5% Aloe vera gel was evaluated by determining… Show more

“…Moreover, both agarose and sericin are used in many biomaterial applications [ 42 , 68 ]. The wound healing property of sericin has been reported in many medical research studies [ 69 , 70 , 71 ]. Sericin increased the attachment of human skin fibroblasts, activated cell migration and proliferation, and activated collagen synthesis, resulting in wound healing acceleration without cytotoxicity [ 72 , 73 , 74 ].…”

“…Sericin increased the attachment of human skin fibroblasts, activated cell migration and proliferation, and activated collagen synthesis, resulting in wound healing acceleration without cytotoxicity [ 72 , 73 , 74 ]. In an in vivo study, sericin gel promoted a faster diabetic wound healing time than bacitracin zinc and polymyxin B [ 71 ]. The wound healing property of sericin in humans was also presented in a study by Siritientong et al [ 75 ].…”

“…In combination with silk fibroin and a poly(vinyl alcohol-based scaffold), collagen was also found to promote cell attachment, spreading and proliferation [ 58 ]. Silk fibroin is a fibrous protein secreted by Bombyx mori silk glands, which exhibits excellent mechanical performance, controllable biodegradability, improved hemostatic profile, great biocompatibility, and low antigenicity [ 59 , 60 ]. Most importantly, in DFUs, it can reduce inflammation and augment the expression of transforming growth factor-β signaling pathway and collagen during wound healing, as reported by Xu et al [ 61 ].…”

“…Additionally, modified with insulin, silk fibroin dressings can also accelerate wound closure, collagen deposition and vascularization [ 62 ]. Sericin is another protein that can be extracted from Bombyx mori silk glands, however, at a lower amount than silk fibroin (80% versus 20% for sericin) [ 59 ]. Silk sericin is a globular protein with remarkable biodegradability, biocompatibility, antioxidant capacity, and regenerative abilities, including faster wound healing abilities, accelerated cell proliferation, and a low inflammatory profile.…”
