Silencing profilin-1 inhibits gastric cancer progressionviaintegrin β1/focal adhesion kinase pathway modulation

Abstract: These findings suggest that PFN1 plays a critical role in gastric carcinoma progression, and these effects are likely mediated through the integrin β1/FAK pathway.

“…In addition, ITGB1 could combine with the FN, subsequently recruit and activate the tyrosine protein kinase FAK, which regulates cell proliferation and apoptosis (Liao, Zhang, Zhao, & Liu, 2018;Park, Ryu, & Han, 2011). Meanwhile, the catalytic subunit of PI3K is transported to the membrane when FAK binds to the SH2 domain of PI3K and catalyse the phosphorylation of AKT (Hwangbo, Kim, Lee, & Lee, 2010), and the ITGB1/FAK/PI3K/Akt pathway is an important signalling pathway involved in cell growth and survival (Cheng et al, 2015). Furthermore, the PI3K/AKT activation is also induced by the E-cadherin in epithelial ovarian cancer cells (De Santis, Miotti, Mazzi, Canevari, & Tomassetti, 2009 Additionally, the ROS level induced by P53 (Li, Dietz, & Harsdorf, 1999) was reduced in the oocyte of 3D culture, which also indicated the oocyte quality was improved.…”

“…It is well known that cumulus expansion has a positive relationship with the quality of oocyte maturation. Cumulus expansion regulated by ERK1/2 (Fan et al, 2009) is a component of the ITGB1/FAK signal pathway (Cheng et al, 2015). Meanwhile, research also shows that the activities of ERK1/2 rely on PCK activities (Prochazka & Nemcova 2019).…”

“…3 Like many other cancer types, initiation and progression of gastric cancer involves multistep processes of various genetic and molecular factors, including activation of multiple oncogenes and suppression of tumor suppressor genes. 4,5 Molecular pathways that affect cell proliferation and migration, including the integrin/focal adhesion kinase, 6 mammalian target of rapamycin (mTOR) signaling, 7 mitogen-activated protein kinases (MAPK), 8 and nuclear factor-jB (NF-jB) pathways, 9 have been demonstrated to play roles in development of gastric cancer.…”
