Silencing of GRP94 expression promotes apoptosis in pancreatic cancer cells

Abstract: Abstract. As a molecular chaperone, GRP94 is the most abundant glycoprotein in the endoplasmic reticulum, playing an important role in maintaining cellular homeostasis. Here, we investigated the expression and the role of GRP94 in regulating cell growth and apoptosis in pancreatic cancer cells. GRP94 mRNA levels were analyzed by QRT-PCR. Immunohistochemistry was performed to localize GRP94 in tissues of the normal pancreas (n=20), chronic pancreatitis (n=20) and pancreatic ductal adenocarcinoma (n=44). Silenci… Show more

“…[136] The cytoskeletal organizational and actin binding proteins like destrin, actin related protein 2/3 complex, subunit 2 (Arp2/3), Neural Wiskott-Aldrich syndrome protein (N-WASP), mitotic kinesin family member 14 (KIF14), Rho GDP dissociation inhibitor β (ARHGDIβ) and microtubule-associated protein (MAPRE2) were all associated with PNI regulation in PDA. [137,138,139,140] Rho/Rho kinase dependent Sema4D/plexin-B1 have been reported to be overexpressed in breast, prostatic and colon cancer PNI. [141] Chemokines CCL5-CCR5 and CXCL12/CXCR4 promote PNI due to increased actin polymerization as a result higher transmembrane Ca 2+ in SACC, breast and prostate cancer respectively.…”

“…In colorectal cancer cells, a high expression of HSP90a has been associated with invasiveness and metastasis (Milicevic et al 2008). The HSP90 isoform GRP94 was reported to partly account for apoptosis resistance in pancreatic cancer cells (Pan et al 2009). …”

“…The most important HSP90 isoforms are the highly inducible HSP90a (HSP90AA) and the constitutively expressed HSP90b (HSP90AB), both of which are mostly found in the cytoplasm and constitute the HSP90A family (Csermely et al 1998, Sreedhar et al 2004, Chen et al 2005. Higher eukaryotes possess their own endoplasmatic reticulum (ER) isoform, glucose-related protein 94 (GRP94; Ni & Lee 2007), which constitutes the HSP90B family and is important for maintaining the ER-homeostasis (Pan et al 2009), as well as their own mitochondrial isoform TNF receptor-associated protein 1 (TRAP1; Csermely et al 1998), which is part of a mitochondrial pathway maintaining mitochondrial integrity (Kang et al 2007).…”
