Silencing cAMP-response Element-binding Protein (CREB) Identifies CYR61 as a Tumor Suppressor Gene in Melanoma

Abstract: Metastatic progression of melanoma is associated with overexpression and activity of cAMP-response element-binding protein (CREB). However, the mechanism by which CREB contributes to tumor progression and metastasis remains unclear. Here, we demonstrate that stably silencing CREB expression in two human metastatic melanoma cell lines, A375SM and C8161-c9, suppresses tumor growth and experimental metastasis. Analysis of cDNA microarrays revealed that CREB silencing leads to increased expression of cysteine-rich… Show more

“…However, the role of CREB in regulating Cyr61 gene expression appears to be tissue specific. In melanoma cells, CREB is a negative regulator of Cyr61 expression (36), while in smooth muscle cells it is a positive regulator (35). In the present study, using deletion analysis, we found that CRE1 and CRE2 were not required for OSM-induced promoter activation.…”

“…Previous reports showed that CREB promoted cancer metastasis (12,14,43,44). CREB regulated vascular endothelial growth factor expression and was involved in human prostate cancer bone metastasis (45).…”

“…CREB does not appear to be primarily responsible for immediate early gene induction in our experiments. Thus, cyr61, which is up-regulated by SRF (19) but down-regulated by CREB (20), is markedly stimulated by serum in an IPMKdependent fashion (Fig. 1A and Fig.…”
