Significant Association of Interleukin4 Intron 3 VNTR Polymorphism with Susceptibility to Gastric Cancer in a South Indian Population from Telangana

Abstract: BackgroundGastric cancer (GC) is the fifth most common malignancy and remains a considerable public health burden worldwide. Genetic variations in genes encoding cytokines and their receptors influence the intensity of the Helicobacter pylori associated inflammatory response, which may contribute to individual differences in the outcome and severity of the disease. Interleukin4 is a typical pleiotropic T helper 2 (Th2) cytokine and is a critical mediator of Th1/Th2 balance. It is involved in the regulation of … Show more

“…Several clinical studies demonstrated that genotype combinations of rs8179190 and other IL-4 polymorphisms significantly influence IL-4 expression (Hussein et al, 2013;Cabantous et al, 2015). In addition, rs8179190 was reported to be associated with susceptibility to chronic inflammatory diseases and cancer (Bhayal et al, 2015;Mohammadoo-Khorasani et al, 2016;Zaaber et al, 2016). We found three studies that assessed the association between IL-4 rs8179190 and CKD susceptibility.…”

“…There are a lot of articles about SNPs of IL-4/IL-13 or their receptors and association with gastric (summary in Supplemental Table S4 [ 45 , 116 , 117 , 118 , 119 , 120 , 121 , 122 , 123 , 124 , 125 , 126 , 127 , 128 , 129 , 130 , 131 , 132 , 133 , 134 , 135 , 136 , 137 , 138 , 139 , 140 , 141 , 142 ]) or colorectal (summary in Supplemental Table S5 [ 62 , 137 , 143 , 144 , 145 , 146 , 147 , 148 , 149 , 150 , 151 , 152 , 153 , 154 , 155 , 156 , 157 , 158 , 159 , 160 , 161 , 162 ]) cancer. Some of them revealed certain kinds of SNPs may serve as a risk factor or prognostic marker for each disease [ 150 ].…”

“…Variations in the IL-4 activity or in the IL-4 receptor due to mutations have been associated with cell proliferation and might affect signal transduction pathways in cancer[ 39 ]. We evaluated INDEL of 70 bp in intron 3 of the IL4 gene (rs79071878), a variation which may influence the production of this cytokine.…”

“…The higher IL-4 production may result in diminished cell-mediated immune response, and escape from immune surveillance in the tumor cells. The cell-mediated immune response may be inhibited by downregulating the expression of Th1 cytokines, decreasing the CD8+ T-cell response in the tumor microenvironment[ 39 - 41 ]. Furthermore, this INDEL has been associated with gastric cancer[ 39 ] and other immune diseases[ 42 , 43 ].…”
