Signalling dynamics in embryonic development

Abstract: In multicellular organisms, cellular behaviour is tightly regulated to allow proper embryonic development and maintenance of adult tissue. A critical component in this control is the communication between cells via signalling pathways, as errors in intercellular communication can induce developmental defects or diseases such as cancer. It has become clear over the last years that signalling is not static but varies in activity over time. Feedback mechanisms present in every signalling pathway lead to diverse d… Show more

“…Around E7.0 for humans and E5.0 in mice, the three lineages (TE, EPI and hypoblast/PrE) of the blastocyst are defined and the embryo is ready to implant in the uterine wall ( Figure 1A ) ( Braude et al, 1988 ; Rossant and Tam, 2017 ). Signaling events regulating early development have been extensively studied over the last decades and have been thoroughly reviewed elsewhere ( Perrimon et al, 2012 ; Menchero et al, 2017 ; Sonnen and Janda, 2021 ). This deep knowledge of the signaling milieu allowed researchers to recapitulate many stages of mouse and human embryonic development in vitro .…”

“…Usually, one group of cells (the inducing tissue) influences the development of another set of cells (the responding tissue) in a process called induction [ 1 ]. Cell-to-cell communication is essential for induction, for the ability of a cell to respond, and to allow dialogue between inducing and responding cells [ 2 ]. Therefore, communication between cells is critical for embryonic development, and the main mechanisms that allow this communication are juxtracrine and paracrine signaling [ 3 ].…”

“…The temporal dynamics of gene expression are known to be a key factor in determining cell fate in many contexts [1][2][3]. Biological and technical advances (e.g., in fluorescent protein reporters and microscopy [4]) have meant that dynamic data has become increasingly abundant (for example, reviewed in [5]), yet dynamic gene expression remains difficult to measure and interpret [6][7][8].…”
