Signaling Scaffold Protein IQGAP1 Interacts with Microtubule Plus-end Tracking Protein SKAP and Links Dynamic Microtubule Plus-end to Steer Cell Migration

Abstract: Background: IQGAP1 is a scaffold protein essential for cellular signaling in response to external cues. Results: Perturbation of the IQGAP1-SKAP interaction results in inhibition of directional cell migration. Conclusion: The IQGAP1-SKAP interaction links to dynamic microtubules at the leading edge to steer cell migration. Significance: IQGAP1 serves as a signaling hub for dynamic interaction between microtubule plus-ends and the cell cortex.

“…SKAP also cooperates with mitotic kinesin CENP-E to govern dynamic KT-MT interaction for faithful chromosome segregation20, which promotes the speculation of CENP-E’s role in translocating SKAP from spindle to kinetochore. Our previous study discovered SKAP forms a cognate complex with IQGAP1 in order to orchestrate directional cell migration, via coupling dynamic microtubule plus-ends, to the cell cortex34. Notably, SKAP functions in different cellular processes while interacting with various proteins.…”

“…29 The enhanced stability of the microtubule cytoskeleton in one particular region of the cell can cause the cytoskeleton to increase growth in that direction. 29 Since the microtubules are the steering wheel of motile cells, 30, 31 increased growth in one region of the cell edge can turn a motile cell. 29 Thus, the local mechanical nature of microtubules within a cell can direct the cell’s motion.…”

“…This was achieved by introducing an 11-amino acid peptide derived from the TAT protein transduction domain into a fusion protein containing amino acids involving binding interface between CSPP1 and CENP-H, as described previously (22,26). Trial experiments were employed to determine the optimal concentration to perturb CSPP1/CENP-H interaction in HeLa cells, which has identified an optimal concentration of TAT-CSPP1-C peptide at 2.5 M (22).…”
