Signaling Initiated by Overexpression of the Fibroblast Growth Factor Receptor-1 Investigated by Mass Spectrometry

Abstract: Overexpression of the fibroblast growth factor receptor-1 (FGFR-1), a prototypic receptor tyrosine kinase, is a feature of several human tumors. In human 293 cells overexpression of the FGFR-1 leads to constitutive activation of the receptor with concomitant sustained high increase in the cellular level of phosphotyrosine-containing proteins. Here we use mass spectrometry to study the tyrosinephosphorylated proteins induced by overexpression of the FGFR-1. Several well known components of FGFR-1 signaling were… Show more

“…Interestingly in addition to known autoantibody targets like prostatic acid phosphatase and PSA, the PCaspecific humoral signature contained proteins that could play a role during tumor development and progression. For instance, the humoral signature included two proteins in the fibroblast growth factor receptor signaling pathway, which has been implicated in prostate cancer development and progression (32)(33)(34). Different predicted molecular phenotypes of increased FGF signaling include increased motility, invasiveness, proliferation, and androgen independence, all of which are ultimately thought to promote tumor progression (34).…”

“…Only recently has mass spectrometry been used to identify many tyrosine phosphorylation sites in receptor tyrosine kinase pathways (15)(16)(17)(18) and to monitor total phosphotyrosine content in pathway proteins over time (19). However, these studies have not provided information on the dynamic regulation of specific tyrosine phosphorylation sites.…”

“…For example, most studies on FGF receptor-mediated signal transduction have been carried out using FGFR1 as the prototypical FGFR, which is bound by many members of FGF family. Others using an overexpression system to characterize the signaling pathways of a given splice variant have had trouble in interpreting the significance of their data, since it is not obtained under physiological conditions (30,31). We previously reported that endometrial stroma expresses predominantly the FGFR2IIIc splice variant by quantitative reverse transcription-PCR analysis and immunohistostaining and that FGF9 potently stimulates endometrial stromal cell proliferation (5).…”
