Sigma receptors: from discovery to highlights of their implications in the cardiovascular system

Abstract: Sigma receptors are the targets of many ligands, of which some (the haloperidol for instance) are psychoactive, and of substances known to have antiarrhythmic effects (amiodarone and clofilium). They are involved in a variety of cardiovascular functions, such as the regulation of cardiac contractility and rhythm and the regulation of coronary and peripheral arterial vasomotricity. This short review will focus on some aspects regarding the ligands, the binding sites, the intracellular coupling and the cardiovas… Show more

“…Such effects may in part be due to activation of receptors on intracardiac neurons. Similarly, various drugs that act on receptors, such as haloperidol, have significant effects on the heart (Monassier and Bousquet, 2002). Our observations raise the possibility that the cardiovascular effects of these compounds may be mediated by their modulation of the electrical activity of intrinsic cardiac neurons.…”

“…28 However, even today the precise role of the receptors is quite ambiguous since they have been implicated in many therapeutic areas such as cardiology, psychiatry, immunology, and oncology. 29 The correlation between castoreum’s affinities to the sigma receptors and its therapeutic effects thus remains obscure at this point.…”

“…-Receptor ligands have profound effects on function in the brain as well as the heart (19,21,35). Progesterone block of -receptors in the brain could play a role in the differences in mood and behavior associated with reproductive state (31,42,46).…”
