Sialyl Lewis x expression in canine malignant mammary tumours: correlation with clinicopathological features and E-Cadherin expression

Abstract: Background: Sialyl Lewis x (sLe x ) antigen is a carbohydrate antigen that is considered not only a marker for cancer but also implicated functionally in the malignant behaviour of cancer cells. Overexpression of sLe x is associated with enhanced progression and metastases of many types of cancer including those of the mammary gland. Canine mammary tumours can invade and give rise to metastases via either lymphatic or blood vessels.

“…34 In dogs, recent reports have confirmed that sialyl Lewis carbohydrates are detectable in canine malignant mammary tumors and that elevated levels of sLe x expression in these cancers are associated with lymph node metastases. 21,27 To our knowledge, this study is the first report of the detection of C2-O-sLe x carbohydrates in canine gastric carcinomas.…”

“…Tissues were counterstained with Mayer's hematoxylin. Incubations with protein block and primary and secondary antibodies were carried out in a humidified chamber at 27 C to 30 C. The remaining staining procedures were performed at room temperature. For comparison with the tumor tissues, samples of stomach from 5 normal dogs were processed and stained similarly using the CHO-131 and Lu-5 antibodies.…”

“…Em pesquisa realizada com carcinomas mamários caninos, observou-se uma relação indireta da expressão das E-caderinas com a de Sialyl-Lewis x (sLe x ), um carboidrato, considerado marcador tumoral, que tem sua expressão aumentada associada à progressão do tumor e desenvolvimento de metástases. Quando a expressão de E-caderinas estava aumentada, a de sLe x estava diminuída, e vice-cersa (PINHO et al, 2007).…”

“…However, the evaluation of LVI and BVI in colon cancer tissue samples by regular HE staining gives significant false positive and false negative results (Merchant et al, 1995;Kikuchi et al, 1995;Nakai et al, 1 , Shao-Yuan Wang 2 2004). Therefore, exploring specific markers for LVI and BVI has become a hot topic of recent years (Pinho et al, 2007).…”
