Shortcut Synthesis of β-Cyclomannin from β-Cyclodextrin

Abstract: Beta-cyclomannin, a cyclic oligosaccharide consisting of seven alpha-D-mannosides connected together by the (1-->4) glycoside linkage, has been efficiently synthesized by the OsO4 oxidation of heptakis(2,3-didehydroxy)-beta-cyclodextrin which was prepared from beta-cyclodextrin by a five-step transformation. The novel cyclooligosaccharide not only showed water solubility high enough to meet the requirement for drug formulation but also demonstrated strong binding ability toward guest molecules. “…The results show that the osmium-catalyzed cis -dihydroxylation of disaccharide derivatives 2,3-dideoxy-4- O -β- d -galactopyranosyl- d - erythro -hex-2-enopyranosides exhibit the same stereospecificity reported for the oxidation of the monosaccharide 2,3-dideoxy- d - erythro -hex-2-enopyranoside congeners: oxidation of 2,3-unsaturated α-glycosides leads to the formation of α-mannosides whereas the oxidation of the respective β-anomers yields β-allosides. 19…”

“…This model was proposed based on the similarity of the anomeric chemical shifts to those of cyclodextrin . A helical MMP structure should have a more‐hydrophilic exterior and a more‐hydrophobic interior than amylose, based on a comparison of cyclodextrins and cyclomannins (cyclic oligosaccharides of α‐(1→4)‐linked mannopyranose) . To test this hypothesis, TROESY NMR measurements were performed on MMP and (MMP+PA) in aqueous solution.…”
